Research Abstract

培養基板の力学特性の空間分布を操作することで細胞の機能や配向をどこまで制御できるか,その可能性を追求するため,表面の薄層のみ力学特性が異なる基板を用いた細胞の基板力学特性検知機構の解明,弾性率が場所により異なる基板を用いた細胞分離,力学的異方性を有する基板を用いた細胞配向制御を進めることを目的として2年間の研究を進めた.研究後半の本年度は,まず,面内で弾性率が変化する基板の作製を行ない,その上での細胞挙動を調べた.既報(Wong et al.Langmuir,2003)を参考にアクリルアミド溶液に光重合開始剤を加え,適当なマスクを当てつつ紫外線照射してゲル化させることで,弾性率が徐々に変化する基板を作製した.弾性率の範囲は既報(Engler et al.Cell,2006)を参考に幹細胞が筋細胞から骨芽細胞に分化する基盤弾性率の範囲である10-50kPaとした.また,基盤弾性率が20kPa違うと別の細胞に分化すると言う報告から,細胞は5kPa程度の弾性率の差を感知できると予想し,細胞長を100μmとした時にその両端で5kPaの弾性率差が生じるように,50kPa/mmの弾性率勾配を持たせることとした.レーザプリンタを用いて適当なグレーレベルのマスクを作製することで,弾性率範囲20~75kPa,弾性率勾配50kPa/mmのゲルの作製ができるようになった.次に作製した基板上で腫瘍由来の血管内皮細胞UV♀2を培養し,その挙動を観察した.その結果,細胞は50kPa付近を境として,より硬い側あるいは柔らかい側に移動する傾向があること,また,硬い領域では細胞が弾性率勾配と直交する方向に動く傾向のあることが観察された.しかし,減光マスクのグレーレベルに応じた微細なパターンがゲル弾性率にそのまま転写され,これが細胞挙動に影響している可能性も否定できず,この点の確認が課題として残された.
