Research Abstract

単細胞シアノバクテリアSynechocystis sp.PCC 6803の走光性運動の細胞間相互作用を顕微鏡観察で評価した。これによって、細胞は低濃度寒天プレート上では個別に運動し、集団を形成しないが、寒天濃度が高いプレートでは集団を形成し、その運動が同期する脈動様の運動を閉めIS田。さらにこの脈動が寒天表面の抵抗を越えたとき、集団全体として走光性運動を示した。また、さまざまな光受容体の変異体も同様の運動特性を示した。このような細胞運動の同期と抵抗の大きい高濃度寒天プレートでの運動能の促進は、線毛による細胞運動の促進とともに、細胞外の刺激が他の細胞の線毛運動の同期を引き起こしたことによると結論された。
糸状性シアノバクテリアArthrospira platensisはらせん形糸状体の回転により運動を示す。この糸状体にサイクリックAMP(cAMP)を添加すると、細胞外多糖は糸状体の全面から分泌が促進され、糸状体の回転によって、後方の末端に蓄積した。この多糖の蓄積が糸状体の物体表面への付着や糸状体同士の付着・凝集を引き起こした。また、cAMPは糸状体の線毛運動によって駆動されると考えられる回転運動を促進した。cAMPは、A.platensisが細胞内で合成し、細胞外へ輸送することが知られていることを考慮すると、液性因子であるcAMPの分泌を介して、線毛運動と細胞外多糖蓄積を促進することで、糸状体凝集という細胞間相互作用を引き起こしていると結論された。
細胞外からcAMPを添加すると、Synechocystisの細胞では約5分で、線毛運動に必要な遺伝子の発現が上昇した。このような遺伝子発現の制御と線毛運動による細胞間相互作用がシアノバクテリアの運動や細胞凝集などの複雑に制御していることが明らかになった。今後は、これらの経路における制御遺伝子の同定が必要と考えられる。
