新規アレルギー治療法の開発に向けた粘膜型マスト細胞の解析

Research Abstract

マスト細胞は存在部位や細胞内分泌顆粒の構成の違いにより結合組織型と粘膜型に分類され、主に前者が生体防御に、後者がアレルギーの発症に関与すると考えられている。アレルギーの予防・治療を考えた際には粘膜型マスト細胞を特異的に標的とする戦略が有効であると考えられるが、粘膜型マスト細胞の分化や活性化機構についてはほとんど解明されていないのが現状である。そこで本研究課題では、免疫学的・生化学的手法、ゲノム・プロテオーム解析を駆使し、未だ実現されていない粘膜型マスト細胞に特異的抗体の樹立と特異的分子の同定、ならびに生体内における粘膜型マスト細胞の分化・活性化機構を明らかにする。本研究課題の2年度にあたる22年度は、初年度に得られた4種類のマスト細胞特異的抗体のうち、1F11抗体を用いた免疫学的解析と生化学的解析を行った。その結果、1F11抗体は腸管や呼吸器に存在するマスト細胞には反応するが、皮膚に存在するマスト細胞には反応しないことを確認し、粘膜型のマスト細胞に特異的であることが判明した。また生化学的解析から1F11抗体は細胞外ATP受容体の一つであるP2X7を認識していることが明らかとなった。なた1F11抗体はin vitroだけではなく、in vivoにおいてもマスト細胞依存的な活性化を抑制できることを見いだした。これらの結果から、今後は、これらの抗体を用いた粘膜型マスト細胞を標的とした抗体療法、免疫療法が確立できるものと期待される。
