Research Abstract

PGC7/Stellaはマウス線維芽細胞において、DNAのメチル化を阻害すること、また、PGC7/Stella遺伝子をマウス線維芽細胞に発現させると、DNAメチル化レベルが低下するだけでなく、高率にトランスフォーメーションが生じることも明らかにしている。本研究では、臓器特異的にPGC7/SEellaを発現させることができるトランスジェニックマウスを作成し、それぞれの臓器におけるがん化や変性疾患の発症を解析することを目的とした。まず、肝臓特異的に発現するアルブミンのプロモーター制御下にPGC7/Stella-IRES-EGFP-polyAを組み込んだトランスジェニックマウス、皮膚特異的に発現するケラチン5のプロモーター制御下にPGC7/Stella-IRES-EGFP-polyAを組み込んだトランスジェニックマウスを作製した。それぞれのマウスにおいて複数のラインが得られ、サザンプロットにより、トランスジーンの挿入が確認できた。しかし、全てのマウスにおいて、EGFPの蛍光およびPGC7/Stellaタンパク質の発現は認められなかった。そこで、よりプロモーター活性の強いCAGプロモーターの制御下に、loxP-neo-polyA-loxP-PGC7-IRES-EGFP-polyAを組み込んだトランスジェニックマウスを作製した。得られたトランスジェニックマウスから、胎児線維芽細胞を採取し、Creリコンビナーゼを発現させることにより、PGC7/Stellaの発現を誘導したが、発現は認められなかった。これらの結果は、残念ながら、何らかの理由によりPGC7/Stellaのin vivoでの強制発現が極めて困難であることを示唆している。
