Research Abstract

前年度に引き続き、"次々世代"の挑戦的萌芽研究:「癌幹細胞の人工的作成」を実施した。前年度までに基盤部分は完成したので本年度は当初の計画に沿ってメカニズム解析を含む先進科学研究を実施し、臨床応用に向けて基盤を構築した。【方法】1)多種の癌細胞で人工癌幹細胞を誘導:腫瘍型に依存するスペクトラムを解明した。未分化100遺伝子全てについて人工癌幹細胞誘導を試行。2)中間体の性状解明。【結果】網羅的オーミックス解析の結果、作成された人工癌幹細胞は細胞表面抗原上で出発点となる癌幹細胞と類似するが複数の相違点があった。類似点は自己複製能や造腫瘍性などの癌幹細胞と共通する特徴を有するが、相違点はcMYC等の癌遺伝子関連転写因子の機能パスウエイに一致するものであり、その時系列的な変化は癌抑制遺伝子(TP53,PTEN)の変異が共存する場合に新たな変異の蓄積が促進され、試験管内での培養操作との関連性が示唆された。現在得られた細胞表面抗原に対する抗体療法等の先進開発に向けて研究継続中である。【重要性】本年度は高精度のメカニズム研究に焦点を絞り、1)未知の発癌分子機構、2)転移・再発・抗癌剤耐性等に関わる悪性化の分子基盤、3)新規分化誘導療法の開発、4)癌の軟骨化等のよる未来医療としての不活性療法等に向けて基盤を構築し展開した。多くの研究分野に強いインパクトを与えると期待される。
