Research Abstract

1.同定した新規軟骨分化誘導剤、永久軟骨再生に必須の遺伝子群のIn vitro軟骨分化誘導能の評価と分化メカニズムの解明
前年度にスクリーニングで同定した軟骨分化誘導剤をin vitroで特異的に肥大化を起さないで軟骨初期分化を誘導することを明らかにした。それらによって、活性化される転写因子(下流のシグナル)を同定するために、軟骨分化誘導剤投与群、コントロールとして非投与群のmRNAの比較をgene chip microarrayを用いて行った。15000遺伝子を搭載したgene chipを用い、2.5倍以上で発現増加、0.4倍以下で発現減少という基準でデータベースを用いた解析を行った。今後は内軟骨性骨化に重要な転写因子をメインに解析を進め、その相互作用を解析し、軟骨分化誘導シグナルの解明につなげる。
2.iPS化したマウス皮膚線維芽細胞の軟骨分化誘導
iPS化に必要な4遺伝子(Sox2,Oct4,c-Myc,Klf4)をレトロウイルスを用いて、マウス皮膚線維芽細胞に遺伝子導入し、多能性幹細胞であることを証明した後に、前年度に同定した軟骨分化誘導剤を添加し、iPS細胞の軟骨分化を検討した。
今後は細胞シート工学を応用した温度感受性培養皿上で軟骨分化誘導剤による軟骨細胞シート作成し、動物モデルを用いて、解析する予定である。
