Research Abstract

本研究では、間葉系細胞の純化・濃縮による再生組織の特性向上を目的として、細胞接着阻害物質MPCポリマーを用いて細胞接着能の違いにより細胞を分取する培養プレート(MPCプレート)を作製し、接着性が高い間葉系細胞を選択的に回収して幹細胞特性の向上(SP細胞濃縮)を図る。最終年度である本年度では、1)MPCプレートで分離したMSCの増殖能、分化能の評価、2)MPCプレートによるSP分画濃縮の有用性検証、をおこなった。1)MPCプレートで分離したMSCの増殖能、分化能の評価、については、前年度の研究で選定されたMPCポリマー濃度5%のプレートを用いて、骨髄細胞を2500細胞/cm^2で播種して継代培養し、一継代10倍程度の良好の増殖を得た。また、多分化能に関しては、MPCプレートを用いて培養したP2,5の細胞と、MPCプレートを用いなかった対照群の細胞を比較検討したところ、MPCプレートで培養した細胞では、骨芽細胞系マーカーであるALPやRUNX2、軟骨細胞系マーカーではCOL2、COL1O、Sox9などの発現が上昇しており、MPCプレートにより、接着性が高い間葉系細胞を選択的に回収でき、幹細胞特性が向上していることが示唆された。2)MPCプレートによるSP分画濃縮の有用性検証、では、MPCプレートを用いて分離したMSCと対照細胞を用いて再生骨を作製した。ラット骨髄細胞をMPCポリマー濃度5%のプレートに播種して細胞を分離し、さらに、10%FBS含有培養液でP2まで継代培養し、人工骨とともにラットの背部に移植した。MPCプレートで分離したMSCを用いた再生骨は、対照細胞を用いて再生骨に比べ、組織学的にも生化学的(アウカリホスファターゼ活性)にも優れ、MPCプレートによる幹細胞特性の向上が検証された。
