Research Abstract

これまでに哺乳類オートファゴソーム形成に関わるAtg因子を多数同定してきた。そこで、本年度はそれらを5つのモジュールに分類してその遺伝学的階層構造を決定した。その結果、ULK1-Atg13-FIP200-Atg101複合体が小胞体の一部にまず局在化し、それに依存してクラスIIIPI3キナーゼ複合体であるAtg14-Beclin 1-Vps34-Vps15がリクルートされることが判明した。その下流では、PI3Pのエフェクターと考えられるDFCP1、WIPI-1、さらに膜伸長因子であると考えられるAtg12-Atg5-Atg16L複合体とLC3-PEがリクルートされる。さらに小胞体膜タンパク質であるVMP1の解析結果を踏まえて、オートファゴソーム形成部位は小胞体の微小ドメインであることを提唱した。
さらにこれらの関連をもとにして、オートファジーの選択的基質であるp62がどのステップでオートファジー関連構造体にリクルートされるかを検討した。従来はp62がオートファゴソーム膜のLC3と直接結合することがその認識機構であると考えられてきたが、今回の解析からp62はLC3を含むほとんどのAtg因子に非依存的にオートファゴソーム形成部位に集積しうることが明らかとなった。さらに哺乳類Atg9もこの部位に一過的に集積することを見いだした。各因子の変異細胞の電子顕微鏡解析を含めながら、オートファゴソーム形成部位の超微形態学的・生化学的解析を進めている。
