Research Abstract

これまで、C型レクチン受容体Mincleが病原性真菌、マラセチアを認識することを見出している。マラセチア真菌は新生児における致死性真菌症の原因菌としても知られており、そのリガンドの同定は重要な課題である。一方、Mincleのリガンドとして、結核菌の細胞壁に存在する糖脂質、トレハロースジミコレート(trehalose dimycolate ; TDM)を昨年同定している。そこで、糖脂質に注目し、様々な溶媒を用いて、マラセチア真菌中に含まれるMincleリガンドの精製、同定を試みた。まず、マラセチア真菌を様々な溶媒で処理し、処理後の菌体のリガンド活性をレポーター細胞を用いて調べたところ、クロロフォルム:メタノール処理で活性は完全に消失し、親水性溶媒処理では影響を受けないことが判明した。このことから、リガンドは脂溶性分子である可能性が示唆された。実際、クロロフォルム:メタノールに抽出されてきた画分をプレートにコートして刺激すると、Mincle発現細胞が強く活性化されることも判明した。この画分を薄層クロマトグラフィー(TLC)で展開したところ、TDMと完全に一致する挙動を示すスポットは検出されなかったことから、TDMとは異なる未知のリガンドを含んでいることが示唆された。そこで、シリカカラムを作成して溶媒の比率を変化させながら分画を進めたところ、両親媒性画分に強い活性を得た。その生化学性質はTDMとは全く異なっており、全く新しい構造のリガンドである可能性が高い。以上の結果より、マラセチア真菌には、Mincleに対する強いリガンド活性を有する新規脂質が存在することが示唆された。
