Research Abstract

本年度は、骨芽細胞性ニッチにおけるニッチ分子に着目し、造血幹細胞の細胞分裂制御に対する機能を明らかにすることを目的に研究を進めた。細胞分裂の解析を進めるにあたり、均一に純化したと考えられる造血幹細胞であっても遺伝子発現にはバリエーションがあり、細胞集団を対象とした解析では、データが平均化され、個々の幹細胞の特性を明らかにすることが出来ないと考えられた。そこで、単一細胞レベルでの遺伝子発現解析により細胞分裂制御機構の解析系(single cell real-time PCR array)を確立した。これまでにマウス骨髄から分離した造血幹細胞(lin-Sca-1+c-Kit+CD48-CD150+細胞)を培養して得られる娘細胞のペア(paired daughter cell: PDC)について、遺伝子の発現解析を行い、Clustering解析およびPartition解析の結果から、p16Ink4a、Ccne1、Ccne2、Ccng2、Cdkn1a、Fbxw7、Foxo1、Foxo4、Tek、Cxcr4、Epcr、Slamf1、Hoxb4、Pbx1、Tert、Notch1、Numb、Bcl2、Lnk、Itga2b、Csf3rなど、21遺伝子がPDCでAsymmetricに発現することを明らかにした。これらの分子は造血幹細胞の細胞分裂様式を制御すると考えられ、今後、これらの分子がいかにして造血幹細胞の細胞分裂制御に関与するのか解析する予定である。
また、人工ニッチ作製に関して、polyethylene glycol(PEG)を用いたmicrowell arrayを作製し、造血幹細胞のsingle cell培養を行う系を確立した。今後、PEG microwell arrayを用いた培養とPDCの遺伝子発現解析を組合せて造血幹細胞の細胞機構を解析し、自己複製を誘導する培養系の確立を図りたい。
