Research Abstract

我々は、世界に先駆けてレプチン-交感神経系による骨代謝の調節機構を見出し、骨代謝調節における新たなパラダイム「神経と骨のネットワーク」を提唱し、骨と骨外臓器は独立して代謝を営んでいるわけではなく、ネットワークを形成し、互いの代謝を調節していることを明らかとした。本研究ではこの概念をさらに発展させ、消化管による骨代謝調節機構、また骨から脳への情報伝達機構を検討することを目的とした。
今年度は、神経系特異的RANK欠損マウスの骨量をマイクロCTにより定量化した。現在、組織学的骨形態計測法により、中枢神経系におけるRANKが骨形成、あるいは骨吸収を調節するのか解析を継続している。また、消化管による骨代謝調節機構について、全身でグレリン受容体欠損マウスが骨代謝の異常を示すことをすでに見出した。そこで、骨形態計測により、グレリン受容体欠損マウスでは骨形成が低下していることを明らかとした。現在、個体レベルにおけるグレリンの骨代謝作用の責任臓器を同定すべく、骨芽細胞、および神経細胞特異的なグレリン受容体欠損マウスを作成し、その解析を行っている。さらに、我々は、中枢神経系や骨で発現する神経ペプチド欠損マウスの骨組織を解析し、骨量に異常をきたす複数の変異動物を同定した。さらに、その一部では中枢神経特異的欠損マウス、骨特異的欠損マウスの作出を行い、個体レベルでの当該神経ペプチドの生理的な骨代謝調節作用の解明を行っている。以上のように、本研究課題は当初計画通り順調に進行している。
