We designed the temperature-responsive and biodegradable block copolymer, poly (N-isopropylacrylamide-co-DL-lactide) (PNIPAAm-co-PLA) was synthesized by ring-opening polymerization of DL-lactide and used as a carrier for an ideal drug delivery system. PNIPAAm exhibits a thermally reversible phase transition in aqueous solution at 32 oC. This transition temperature is called the lower critical solution temperature (LCST). When the temperature is below the LCST of the thermo-responsive block, nanoparticles are formed with the hydrated outer shell and the hydrophobic inner core. When the temperature exceeds the LCST, the outer shell shrinks and becomes hydrophobic. Our temperature-responsive nanoparticles successfully exhibited thermo-responsive drug release and cellular uptake while possessing biodegradable character.
