In osteoclastgenesis system using human peripheral-blood mononuclear cells, we found that calpastatin was cut off and calpain revitalization was elevated from 3 day to 6 day. The protein level changes of calpain 1 and calpain 2 were not detected in our osteoclastgenesis system. Cell permeable calpastatin down-regulated the osteoclastgenesis and the bone absorptivity of human osteoclast. After 15 minutes TNFα (100ng/ml) stimulation, calpain revitalization elevated in human osteoclast.
