Establishing methods to prepare high quality Islets of Langerhans for transplantation is an important subject directly linked to improving transplantation success. This research aims to prevent hypoxic damage to the Islets of Langerhans. We accomplished this by introducing a method to ensure consistent oxygenation from pancreatic organ storage to the completion of pancreatic enzyme treatment. The method involved injecting artificial hemoglobin into the pancreatic ducts to oxygenate the pancreatic tissue during storage. The introduction of oxygenation to the pancreatic ducts via artificial oxygen carriers, as well as the use of hollow fiber pressurization modules within the donor islets clearly improved the output, energy status, and viability of islets and suppressed apoptosis.
