Research Abstract

RNAの化学合成において、2'-ヒドロキシ基の保護はその成否を握る。これまでに様々な保護基が合成されてきたが、真に有効な保護基は未確立の状況にある。本研究では、構造が単純であり酸・塩基に強いアリル基に着目した。我々が開発したカチオン性CpRu錯体と2-キノリンカルボン酸との混合触媒は、メタノール溶媒中、アリルエーテルを定量的に脱保護できる。その他の添加剤は不要であり、アルコールとの共生成物も揮発性アリルメチルエーテルなので、目的物の単離効率は高い。ウリジン三量体(3-5U)を標的化合物に定め、2'O-およびリン酸部にアリル保護基を導入した。これを、メタノール中、CpRu触媒で処理することにより、定量的にアリル基を除去することができた。残る問題は触媒と生成物との分離にある。如何に触媒を除くかは有機合成効率の高低に大きく影響する。本触媒を「高分散性・低保磁力・高飽和磁化率」を特徴とする、Fe_3O_4磁性粒子を含有する数μmの球形マイクロSiO_2粒子に担持し、操作性を向上するとともに、触媒の分離、回収を可能とした。上記アリル保護トリリボヌクレオチド基質を先の不均一系触媒を用いて脱アリル化、(C_2H_5)_2NH処理後、定量的に3-5Uのアンモニウム塩を得ることができた。均一系では単離収率が低下する傾向にあるが、本不均一系触媒を用いた場合には脱アリル化/ジエチルアミン添加/磁石による触媒分離/溶媒留去の過程での生成物の損失はなく、単純操作でほぼ定量的単離が可能となった。これら結果はRNA合成におけるアリル基の潜在的有用性を示すものである。
