Outline of Research at the Start

乳酸菌が生産する抗菌ペプチド・バクテリオシンは、低濃度で強力な抗菌活性を有し、安全な抗菌物質として期待される。これまでに特徴的な構造と活性をもつ多種多様な新奇乳酸菌バクテリオシンを見出し、これらの新しい生合成・作用機構を明らかにしつつある。本研究では、特異な構造と活性をもつものをさらに探索するとともに生合成・作用機構を明らかにし、新奇バクテリオシン群の実用と生合成機構を利用した新奇抗菌ペプチドの創出に向けた分子基盤の確立を目指す。

Outline of Final Research Achievements

From lactic acid bacteria isolated from diverse sources, many novel bacteriocins characteristic in structure and activity, especially various class II bacteriocins and class III bacteriocins (antimicrobial proteins), were identified. Biosynthetic gene clusters for novel bacteriocins including circular and glycosylated bacteriocins were determined and characterized for self-immunity, secretion, and maturation. A multiple bacteriocin transporter were demonstrated for its availability in secretion of various antimicrobial peptides.

Academic Significance and Societal Importance of the Research Achievements

安全な抗菌物質としての様々な用途での利用が期待される乳酸菌バクテリオシンについて、新しい生産株や新しい構造を有するものを多数見出した。また、種々の新奇乳酸菌バクテリオシンが有する特徴的な生合成機構や作用機構の一端を明らかにした。さらには、乳酸菌バクテリオシンが有する生合成機構を利用した、抗菌ペプチドの分泌発現、評価系の基盤を構築した。これらの成果により、用途に応じた乳酸菌バクテリオシンの選択が可能となり、より効果的な抗菌ペプチドの創出への展開が期待される。
