Outline of Research at the Start

代表者らはクロマチンの集合体が、従来広く描かれていたようなクロマチン線維がループ状に束ねられたものではなく、コンパクトな塊のようなドメインであるという仮説を提唱している。本研究課題では、ユークロマチン領域を蛍光修飾し、修飾領域のヌクレオソームの新規1分子イメージング、計算機シミュレーションを組み合わせ、クロマチンの集合体のなかのヌクレオソームの動きを可視化し、その動きを解析することでこの仮説を検証する。さらにこのドメインが液滴様構造体であるかも合わせて検証する。

Outline of Annual Research Achievements

代表者らはヒト生細胞における200kb程度のクロマチンの集合体が、Hi-C解析をもとにしばしば描かれるクロマチン線維がループ状に束ねられたものではなく、ループを構成する線維がコンパクトな塊(ドメイン)を作っているという仮説を提唱している(Maeshima et al.,Curr.Opin. Cel l. Biol. 2020など)。塊であると転写因子などのタンパク質の局所濃度が増加し、反応の協同性や効率化など、新たな制御が可能となる。本課題では生細胞におけるクロマチンの構造情報を得るために、近接した2つのヌクレオソームを蛍光標識し、その2つの動きを超解像で可視化、追跡する 新規システムを確立する。そして、その2つのヌクレオソームの動きの空間相関を解析することでこの仮説を検証する。

令和4年度はコンパクトな塊のようなクロマチン集合体仮説の検証をおこなった。令和3年度で確立された手法を用いて、数千個の近接したヌクレオソームペアの動きのデータを蓄積した。統計解析により、どのくらい近接し たペアが空間相関を示すか(または示さないか)を明らかにした。その結果、ヌクレオソームの距離が集合体サイズ程度(150 nm)以内で、両者の動きが高い空間相関 (相関係数0.6以上)を持つことが分かった。以上のことから、クロマチンの集合体はコンパクトな塊であることが明らかとなった。

Reason

当初の計画通りに統計解析により、どのくらい近接し たペアが空間相関を示すか(または示さないか)を明らかにし、ヌクレオソームの距離が集合体サイズ程度(150 nm)以内で、両者の動きが高い空間相関 (相関係数0.6以上)を持つため、クロマチンの集合体はコンパクトな塊であることを明らかにしたため。

Strategy for Future Research Activity

計画は概ね順調に遂行できたため、さらに大量に画像デー タを得て、近接した緑・赤色で標識されたヌクレオソームのペアを抽出し、それぞれの動きを追跡する。そして得られた結論の確認を高精度におこなう。
