Outline of Research at the Start

miRNAは約22塩基程度の小さなnon-coding RNAであり、相補的な塩基配列をもつmRNAに対合し、RNAサイレンシングという機構により翻訳を抑制する。miRNAは多数の遺伝子を一括して抑制するだけでなく、その程度もさまざまという非常に複雑な遺伝子発現調節をおこなうことから、記憶・学習・思考などの高次脳機能の分子メカニズム解明の鍵となる分子と考えられる。しかしながら、その遺伝子発現制御機構の複雑さから、どのように高次生命機能を制御しているのか、詳細は不明である。そこで、本研究ではさまざまな手法を組み合わせ、miRNAが制御する複雑な遺伝子ネットワークの詳細を明らかにすることをめざす。

Outline of Annual Research Achievements

ヒトには2,000以上のmicroRNA(miRNA)という小さなノンコーディングRNAが存在し、それぞれが複数の遺伝子の発現を異なる程度で微調整していることが近年のRNA研究によりわかってきた。このような極めて複雑な遺伝子発現の制御機序は、特に学習・記憶・思考などの高次脳機能の調節や、ヒトに特有の生体防御機構などの複雑な生命機能を制御するのに適したシステムと考えられる。しかしながら、従来の研究ではmiRNAを介した高次脳機能における遺伝子ネットワークの詳細は明らかになっていない。本研究では、トランスクリプトーム解析と共に、独自性が高く、従来法とは異なる視点からの実験的解析手法と、申請者らが新たに開発したバイオインフォマティクス的解析手法を組み合わせ、高次生命機能におけるmiRNAによって構築される遺伝子ネットワークの詳細を明らかにし、その動作原理の解明をめざしている。具体的には、miRNAの生合成過程の制御機構、miRNAに優先的に結合する二本鎖RNA結合タンパク質の種類とその発現制御機構、さらにはmiRNA-mRNAのネットワーク解析により、複雑な高次生命機能の制御機構を解明する。

Reason

miRNAは、それぞれが異なる複数の遺伝子の発現を異なる程度で制御している。このように、miRNAは遺伝子発現を複雑に微調整できることから、特にヒトに特有な生体防御機構や高次脳機能の調節に適したシステムと考えられる。しかし、従来の研究では、そのような複雑な生命現象におけるmiRNAを介した遺伝子ネットワークの詳細は明らかにされていない。本研究では、実験的研究手法と計算科学的研究手法の両方を用いて、miRNAによる高次生命機能の制御機構を解明することを目指している。申請者らは、特定のmiRNA群の活性は、それらに特徴的な二次構造を識別して結合する二本鎖RNA結合タンパク質によって制御される可能性について検討し、実際に二本鎖RNA結合タンパク質の種類によって、miRNAのプロセシングのステップが制御されることを明らかにした。さらに、その下流のmRNAの発現解析を行うことで、成熟型miRNAによるmiRNA-mRNAネットワークを解析した。一方で、二本鎖RNA結合タンパク質同志の相互作用についても、共同研究によるマススペクトロメトリー解析と計算科学的研究手法により明らかにした。その結果、miRNAの成熟過程に関わるタンパク質と相互作用する二本鎖RNA結合タンパク質同志の相互作用により、miRNAの成熟過程が制御されることがわかってきた。さらに、制御されるmiRNAの種類は外部からの刺激による二本鎖RNA結合タンパク質の発現変動により、巧みに制御されていると考えられる結果が得られた。

Strategy for Future Research Activity

高次生命機能におけるmiRNA-mRNAによる遺伝子ネットワークの制御は、外部からの環境要因によって変動する可能性が強く示唆された。それは、特に二本鎖RNA結合タンパク質の発現変動が大きな影響をもたらすと考えられた。今回の研究結果から、二本鎖RNA結合タンパク質同志の相互作用がmiRNAの成熟化過程に大きな影響を与える可能性が提唱できたので、この点について新たな検討を行う。
また、miR-124aはヒト・マウス・ラットで塩基配列が同一で、脳に発現する全miRNAの25~48%を占める脳でもっとも多く存在するmiRNAである。そのノックアウトマウスでは脳の発達障害や「記憶」に重要な海馬の神経回路形成に異常が認められるため、ニューロンの維持・機能に関与するmiRNAと考えられている。miR-124aによる遺伝子ネットワークは解析できていないが、miR-124aの二本鎖RNA結合タンパク質による制御機構についても検討する予定である。
