Outline of Research at the Start

Outline of Final Research Achievements
In this study, we performed long-read whole genome sequencing (WGS) on multiplex families with autism spectrum disorder (ASD), which are expected to efficiently facilitate the identification of ASD susceptibility variants, to investigate the mechanisms underlying ASD pathogenesis. Given the high cost of long-read sequencing, we first calculated polygenic risk scores (PRS) based on genome-wide genotyping data for ASD, in order to assess the contribution of common variants to ASD in multiplex families. Based on these results, we selected families with a high likelihood of harboring ASD susceptibility variants for long-read sequencing. In parallel, we developed a pipeline for the detection of small-scale copy number variants (CNVs) and repeat expansions from long-read data, and conducted analyses to identify such small-scale CNVs.

Academic Significance and Societal Importance of the Research Achievements
