| Outline of Research at the Start |
がん細胞特異的な遺伝子変異に由来する抗原（ネオアンチゲン）は強い免疫反応を誘導する。本研究では、進行がん患者にも臨床応用できるように、3次元培養（オルガノイド）した腫瘍組織から最先端の質量分析（ペプチドミクス解析）技術を用いて、ネオアンチゲンを効率的に同定する手法を確立する。また、同定したネオアンチゲン特異的T細胞の抗腫瘍効果を確認する。本研究の成果は、“がんの免疫学的病態の解明”に寄与するとともに、“ネオアンチゲンを標的とした個別化がん免疫治療”の臨床開発を加速するものと期待される。

| Outline of Final Research Achievements |
No reliable methods have been established to identify gene mutation-derived antigens (neoantigens) that play a central role in anti-tumor immune responses. In this study, we developed a method to efficiently and comprehensively identify neoantigen peptides presented on HLA molecules on the surface of cancer cells by mass spectrometry (peptidomic analysis) using tumor organoid cultures. In addition, tumor-infiltrating lymphocytes isolated from tumor tissues were examined for their reactivity to organoid tissues and neoantigens.

| Academic Significance and Societal Importance of the Research Achievements |
がん免疫療法開発において、がん抗原、特にネオアンチゲンの同定効率の低さが課題とされている。本研究では、微量の腫瘍組織から拡大培養したオルガノイド組織を用いて、ネオアンチゲンを効率的・網羅的に同定する手法を開発したが、その成果は“がんの免疫学的病態の解明”に寄与するとともに“ネオアンチゲンを標的とした個別化がん免疫治療（がんワクチンやT細胞受容体遺伝子導入T細胞療法）”の開発を推進するものと期待される。
