Outline of Research at the Start

歯周病やインプラント周囲炎による歯槽骨破壊は自己免疫疾患としての性質を呈しているが，詳細な破壊機序は解明されていない．我々はストラスブール大学のミュラー教授との共同研究により，自己免疫疾患におけるオートファジー異常を正常化させることで免疫トレランスを誘導する新規ペプチド(P140)を応用し，歯周病マウスの歯槽骨破壊抑制に成功した.　これは，歯周病がオートファジー関連疾患である可能性を示唆している.　本研究では歯周病やインプラント周囲炎における歯槽骨破壊を，炎症局所のマクロファージ (Mφ)活性に注目し，オートファジー異常の観点から病態を解明する．

Outline of Annual Research Achievements

本研究は，歯周病やインプラント周囲炎における病態形成メカニズムを，歯槽骨破壊局所に集積するMφの活性化に注目し，オートファジー異 常の観点から解明するとともに，炎症性サイトカインの産生経路を特定し，免疫トレランス獲得につながる新たな検査技術や 新規組織再生療 法開発につなげることを目的としている.　本年度はマウス歯周病モデルに対してオートファジー正常化ペプチドであるP140を腹腔内投与し，誘導後10日目に屠殺・サンプルを回収，顎骨由来マクロファージを回収しオートファジー関連遺伝子の変化を確認したところ，Ulk-1, P62の遺伝子発現が歯周病誘導群と比較して有意に抑制されていることがわかった．また，オートファジーエンハンサーを培養液に添加することで強制的にオートファジー活性を促進したところ，炎症性サイトカインの一つであるTNF-aの遺伝子発現が亢進し，オートファジー関連遺伝子と炎症性サイトカイン遺伝子の発現に関連がある可能性が示唆された．
