Outline of Research at the Start

これまでの神経科学研究では、外側膝状体などの神経核における下位領域や大脳視覚野における特定の領野群において、空間視覚情報の処理に関わる神経活動が観測されることが分かってきた。しかし、ヒトにおいて空間視覚情報処理に関わる脳領域の機能と構造がどのような関わりを持つかについては未だに明らかになっていない。 本研究提案では、定量的MRI・機能的MRI・拡散強調MRI・心理物理実験という4つの手法を組み合わせることで、ヒト脳における空間視覚情報処理の神経機構を構造・機能の両面から包括的に評価する手法を確立させ、ヒトの空間視覚情報処理機能の神経基盤を検証する。

Outline of Annual Research Achievements

Macromolecular Tissue Volume法(Mezer et al., 2013)による定量MRI計測を用いて、外側膝状体の下位領域を構造データのみから同定する研究を進めた。具体的には、test-retest reliabilityの分析、inter-rater consistencyの分析、部分容積効果の影響を評価する分析を進めることで、得られた外側膝状体下位領域分析結果の信頼性および妥当性を示した。さらに、定量MRI計測値が大脳基底核における既知の解剖学的な違いを検出できていることを示すことで、計測データの妥当性を示した。加えて機能的MRIデータと定量MRIデータを比較することで、定量MRIで求められた外側膝状体の下位領域は、先行する侵襲的な電気生理学研究の知見と一致した刺激選択性を示すことも明らかになった。この一連の成果は、NeuroImage誌に原著論文として掲載されるに至った(Oishi et al., 2023)。

また機能的MRIと拡散強調MRIの統合的な分析により初期視覚系の脳領域間の構造共変性および遺伝率を検証した結果について、Journal of Neuroscience誌に原著論文として発表した(Miyata et al., 2022)。

このほか、機能的MRIと構造MRIデータの統合的分析の性能を畳み込みニューラルネットワークを用いて向上させる試みを開始し、国際会議での発表を行なった(Benson et al., 2022 Vision Sciences Society)。一連の研究成果について、国内の研究会(第71回脳の医学・生物学研究会)で招待講演を行なった。国際会議Organization for Human Brain Mappingにおいて神経解剖学と脳イメージングの融合に関する教育コースを主催し、自身の外側膝状体に関する研究成果を含む脳の構造・機能連関についての教育講演を行なった(Takemura, 2022 Organization for Human Brain Mapping)。

Reason

当初計画していた定量的MRIによる外側膝状体下位領域を対象とした研究が無事に原著論文として出版されたほか、機能的MRIと構造MRIを組み合わせることによる新たな研究成果も得られているため、本研究は順調に進捗していると考えられる。

Strategy for Future Research Activity

外側膝状体の下位領域のマッピングについて、両領域の解剖学的な違いをさらに理解するために、MRIデータのみならず死後脳から得られた解剖学データの分析も進めていく。視覚系の白質線維束の分析に有用な拡散強調MRI計測のパルスシーケンスの検討を行う。fMRIで同定された空間視覚情報処理に関わる脳領域間が、どのような白質線維束を介して連絡をしているのかを拡散強調MRIデータの分析によって明らかにする研究を実施する。空間視覚情報処理の個人差を評価する心理物理実験を実施する。
