Outline of Research at the Start

脳オルガノイドは、iPS細胞など多能性幹細胞が自己組織的に３次元構造化した多細胞塊であり、発生や進化などの学術研究、再生医療や病態研究などの応用研究に広く活用されている。しかしながら小脳オルガノイドの３次元構造は、いまだ胚子期の組織構築に留まり、小脳皮質形成の複雑な発生過程を十分に再現するには至らない。本課題では「多能性幹細胞の自己組織化能の誘導と細胞のアセンブルにより高度な小脳オルガノイドモデルを創出し、脳科学における新たなヒト脳研究分野を創出すること」を目的とする。ヒト脳研究に資する新たなモデルとして開発し、神経回路形成、組織破綻、機能不全とその修復機構の解明に寄与することを目指す。

Outline of Annual Research Achievements

脳オルガノイドは、iPS細胞など多能性幹細胞が自己組織的に三次元構造化した多細胞集団であり、発生過程を模倣した脳組織モデルとして、発生・進化などの学術研究から再生医療や病態研究などの応用研究まで幅広く活用されている。例えば、大脳オルガノイドでは神経発生で見られる神経上皮の形成、細胞分裂、細胞周期に伴う核移動（エレベーター運動）、inside-out様式の細胞移動、層特異的な分子マーカーの発現など、一連の経時的な素過程が再現され、大脳皮質特有の明瞭な層構造が再現される。しかしながら、小脳オルガノイドにおいてはそのような明瞭な層構造・組織形成がまだ不十分であり、現在の培養技術で再現できる構造は、胚子期後期相当の組織構築に留まる。これは層構造の形成過程が大脳と小脳では全く異なることに起因していると考えられる。大脳皮質では組織構築に関わるほぼ全ての神経細胞が脳室帯から産生されるのに対し、小脳では異なる二つの胚芽領域から生み出され、複雑な細胞移動を経て層構造が形成される。本課題では、多能性幹細胞の自己組織化能の誘導と細胞のアセンブルを組合せ、より高度な小脳オルガノイドモデルを創出することを目指した。分化条件の精緻化によりプルキンエ細胞に代表される脳室帯由来細胞の分化効率を既報の3倍以上に高めることに成功した。非神経細胞を別途作成し、これらをアセンブルすることでより複雑な組織構築を伴う小脳オルガノイドを作成した。さらに共焦点顕微鏡によるオルガノイドのライブイメージングシステムと、レポーター遺伝子（ATOH1-tdTomato, KIRREL2-Venus）ノックインiPS細胞の利用により、個々の細胞の挙動を定量的に解析した。改良型小脳オルガノイド由来のプルキンエ細胞は疾患モデルマウスへの移植により、細胞機能の発現を一部確認することに成功した。
