Outline of Research at the Start

本研究では,地球外物質によって原始地球上に供給された分子が原始地球上での生命誕生の材料となったという仮説について,とくに遺伝物質である核酸生成への地球外物質(彗星,炭素質隕石,惑星間塵:いずれも原始地球上への主要な有機物供給源)の寄与を,模擬彗星サンプルと隕石有機物の分析で明らかにする。
彗星に含まれる分子を模擬実験で作製し,反応生成物を模擬彗星サンプルとして扱うこととする。隕石有機物は現有の炭素質隕石から抽出する。
対象化合物は液体もしくはガスクロマトグラフ/質量分析計で分析する。

Outline of Annual Research Achievements

代表的な炭素質隕石であるマーチソン隕石から,生命の遺伝子の構成成分,核酸塩基を効率的に抽出するために,マーチソン隕石粉末から塩酸で煮出す手法を開発した。110℃の6M塩酸で12時間加熱し,上澄みを回収した。回収した上澄みは陽イオン交換カラムクロマトグラフィ-で分画され,無機金属イオンなど夾雑物が除去された。生成した抽出液は液体クロマトグラフ―超高分解能質量分析計で分析され,シトシン,ウラシル,アデニン,グアニン,チミンすべてが検出された。検出濃度は水のみを用いた抽出よりも数倍高く,本手法が微量地球外サンプルからの抽出法として理想的だとわかった。
2023年9月に小惑星ベヌーから回収されたサンプルが地球に帰還し,同サンプル中核酸塩基を分析する前に,回収サンプルの立体構造分析等での利用が検討されている,X線トモグラフィーが試料中有機物にどのような影響を与えるか,実際の炭素質隕石を用いて検証した。均一に粉末化したマーチソン隕石を等分し,一方に~180GyのX線を暴露し,もう一方は比較用として同操作はおこなわなかった。これら2種の隕石に含まれる核酸塩基を分析すると,それらの種類や存在量に大きな違いは見られず(5%以内),X線暴露の影響は無視できることが明らかになった。
小惑星ベヌーで回収されたサンプルの予備分析で,窒素を含む有機化合物が多いことがわかり,本来予定になかったサンプル(~18mg)が我々の分析チームに配分された。上記分析法を駆使し,ベヌーサンプル中から核酸塩基を含む多様な窒素複素環化合物を検出することに成功した。

当初の予定では23年度中の分析は不可能と思われた小惑星ベヌーサンプルが配分され,大きな問題点なく分析が遂行できたため。

Strategy for Future Research Activity

小惑星ベヌーサンプルに含まれる核酸塩基など窒素複素環化合物を網羅的に解析する。さらに,24年夏以降に予定されているサンプル配分(~0.6g)の本番を見据え,分析環境の再確認,およびサンプル到着後の分析を効率的に遂行する。分析結果と小惑星リュウグウ・炭素質隕石分析結果や模擬実験結果と比較し,地球外物質中核酸塩基の生成メカニズム解明に資する。成果を速やかに論文にまとめ,準備が整い次第公表する。
