Outline of Research at the Start

これまでに,適切にデザインした自己組織系膜場で,特異的な機能(例 分子認識/変換機能)を誘導・制御できる事を明らかにしており,自己組織化膜構造体を固定化した分離担体(Membrane-on-Membrane)を設計開発してきた.両者を【融合】した革新的な分離技術を確立し,ケーススタディとしてプロセス開発への応用の可能性を検討する.
特に,両親媒性触媒/分子(4級アンモニウム系触媒, 脂肪酸系界面活性剤など)を,(1)液液界面(油相-水相系),ならびに,(2)固液界面(cryoゲル), (3) Cubosomalゲルに機能化し,高度な分子変換能・分子認識能を誘起・制御する手法を明らかにする.

Outline of Annual Research Achievements

本申請課題は、従来の化学プロセスで活用される「異相系」の界面を、分子が秩序高く配向する「自己組織系」に置き換えることにより、エンタルピー駆動型プロセスをエントロピー駆動型プロセスに変革することを大目的とする。2023年度は、(i)「自己組織系」界面の物性評価、ならびに、(ii)「固液系-自己組織系融合材料」の開発の重点課題のうち、特に(i)に注力して検討を進めた。前者は、時間分解蛍光スペクトル解析手法(TRES、後に、Decay-Associate Spectroscopy法(DAS法)に改称)の開発について検討し、学術論文として成果を公開した。従来から多用されるベシクル型自己組織系のみならず、Cubosome、Cataniosomeなどの新規材料、さらに、(ii)で開発した固相-自己組織
系融合材料の表面特性解析ツールとして有効である事を明らかにした。(ii)は、2種類の担体を対象にして検討した。その一つであるCubosomalゲルについては、ゲル表層を各種両親媒性界面活性剤(ハイドロコルチゾンほか)で修飾することに成功し、成果報告した。また、前年度に確立した多孔性高分子材料の調製方法(逆ライデンフロスト・クライオゲル法)を活用して、天然資源からなる、同様の物性(メソ-マクロ孔、柔軟性、可逆性、ほか)を有するポリマー粒子を合成することに成功した。さらには、リン脂質2分子膜を被覆して、(i)の方法で物性解析を検討した結果、マクロ的な物性の変化(例 圧密変化、伸縮変化など)に応じて、メソスケール物性の変化(例 疎水性、水和状態など)を惹起できるメカノケミカル制御可能な新規な分離担体が設計開発可能であることを示している。予定通り、研究が進捗している、副産物として、インパクトの高い分析手法(DAS法)など、他にも転用可能な基盤技術を確立することに成功した。

2: Research has progressed on the whole more than it was originally planned.

Reason

当初計画に記載した、(i)自己組織系界面の解析手法の確立、および、(ii)固液系-自己組織系融合担体の調製方法の確立、いずれも、順調に研究データを取得している。(i)については、複数の学術論文として公表した(DAS解析手法)。(ii)については、前年度に確立したポリマー担体調整法を他の天然ポリマー型素材の開発にお応用することに成功している。また、その後の検討の結果、脂質膜被覆単体のメカノケミカル制御性を明らかにしている。両者をつなぐために、現在、マイクロ流体デバイスを活用したマイクロプロセスの設計開発に着手している。

Strategy for Future Research Activity

現時点では、特に問題は発生しておらず、当初予定通り、研究計画を推進する。実際に、固液系-自己組織系融合材料を調製する手法、また、メソ-マクロ物性を解析して制御する手法は既に確立し、基礎データを蓄積しつつある。2023年度で展開した材料や方法論は、計画当初の想定を上回るパフォーマンスを示す可能性があり、特定ケーススタディとして研究を進めている。客観的な視点・思考を忘れずに、着実にデータ取得に専念したい。2023年度では上記の上位概念をターゲットに入れながら、当初計画通り、工業利用可能な材料としての可能性を検討するための検討を、着実に進めていく。その前段階として、マイクロ流路デバイスと開発した材料の複合化によるマイクロプロセスの設計開発に着手しており、現在、基礎的なデータの集積に従事している。
