Outline of Research at the Start

代表者らが開発した独自の新たな円偏光変調手法による,先端的な高感度・高確度円偏光二色性(CD)顕微イメージングに対し,検出感度のさらなる向上,観察波長域の紫外域への拡大,ライブイメージングの実現等を目指した研究を行う。これにより高度化・拡張・汎用化したCD顕微イメージングの手法を,キラルな微結晶や分子会合体などの光学特性解明,掌性同定などに適用し,キラル光学イメージングによってのみ可能な新手法を開拓・確立する。またライブイメージングにより,キラル構造の生成プロセスの観察手法,生物試料への適用により,キラル高次構造を持つ分子種の分布・動態観測を目指した手法開発を行う。

Outline of Annual Research Achievements

昨年度に引き続き,高繰り返しレーザーを用いた高速円偏光変調方式によるCD顕微イメージングの有効性確認に向けた実験を進めるとともに,CDイメージングの高速化を目指し,類似の円偏光変調方式とイメージセンサーを用いたCD顕微イメージング装置の開発を行った。イメージセンサー方式では,円偏光変調した光を試料に照射し,そのイメージをイメージセンサーで観測し,復調することでCDイメージを得る方式を試みている。現時点で,何らかの要因で信号に系統誤差が入り,高感度でCD信号を得ることがまだできていないが,その要因が判明しつつあり,次年度にはこの方式によるCDイメージングが可能になると期待している。
また本年度は,深紫外領域におけるコヒーレント光を用いた円二色性イメージング計測の実現に向けた技術開発をさらに進めた。イメージングを実現するために必要となる,深紫外コヒーレント光源を用いた顕微イメージング計測系を構築することに成功した。また,開発した顕微イメージング系の検出器をピンホールと光電子増倍管に交換した場合に,円二色性が検出可能であることを確認することに成功した。さらに,円二色性イメージングの実現に向けた試料スキャンシステムの構築を開始した。
近接場領域の高空間分解能で円偏光二色性イメージングを実施する新たなアプローチとして,原子間力顕微鏡を基盤とした光誘起力顕微鏡を用い,励起光に左右円偏光を導入することでCDイメージを得る実験研究を進め,2次元金属ナノ構造試料でその有効性を確認し,論文の掲載に至った。

Reason

イメージセンサーを用いたCD顕微装置の開発を進め,その問題点が明らかになってきており,この点で想定した進展が得られていると考えられる。また光誘起力顕微鏡による近接場CDイメージング手法の開発は,その可能性を示して論文公表にまで至っており,想定以上の進展があったと考えている。さらに,深紫外領域における円二色性イメージングの実現に向けて不可欠となる,対物レンズ,ピンホール,光電子増倍管を用いた顕微円二色性計測系の構築に成功したことは,目標の達成に向けた大きな進捗である。一方で,分担者1名の分子科学研究所からの転出を埋める研究者の募集が想定通りに進まず,年度末にようやく特任助教の採用に至った状況で,この点では計画推進が十分進んでいるとはいえない面もある。次年度には採用した特任助教が力を発揮して本課題が大きく進展することを期待している。

Strategy for Future Research Activity

計画に従い,光学チョッパの改良,試料スキャン方法の改良,反射型配置法の試験の推進,発光励起円偏光二色性検出法の開発推進,測定波長域の拡張等を引き続き行う。またイメージセンサーを用いたCD顕微イメージング装置の構築を継続して進め,その特性評価を行う。同時に,様々なキラル構造ナノ・マイクロ物質の円偏光二色性イメージング計測を,共同研究を含めて行い,装置のポテンシャルを検証する。光誘起顕微鏡による円偏光二色性イメージングについても,装置の高感度化・高精度化を推進するとともに,ナノ構造試料の測定を通じて,その有効性の評価を進める。
深紫外領域における円二色性イメージングの実現に向けて,測定用試料を高精度にスキャンするための系を構築する。昨年度までに準備した深紫外円二色性顕微計測系と融合することにより,深紫外円二色性イメージング計測を実現する。
