Outline of Research at the Start

ゲルやスポンジなどの含水性高分子網目材料は、生体組織と類似の構造・物性を有し、材料科学的にも生体材料応用を目指す上でも興味深い材料である。その一方で、両者には、網目サイズや内包する水との相溶性など、相違点も多く存在する。
本研究では、「ゲルとスポンジの類似性と特異性」を明らかにし、ゲルとスポンジを繋ぐ横断的な学理を確立することを目的とする。申請者が世界で初めて確立した、同一の成分組成からゲルとスポンジを作り分ける手法を用い、ゲル、スポンジ、ゲル・スポンジ中間物質を系統的に作製し、構造・物性を網羅的に測定・解析することで分野横断的な体系化を試みる。

Outline of Annual Research Achievements

生体組織に類似した特性を有するゲルやスポンジなどの含水性高分子網目材料は、その応用可能性から有望なバイオマテリアルとして期待されている。しかし、これらの材料間の科学的枠組みは未だ確立されていない。本研究では、ゲルとスポンジの相違点と共通点を徹底的に探求し、両者を包括的に理解するための統一理論を構築することで、双方を隔てるギャップを埋めることを目的としている。
2023年度の研究では、ゲルとスポンジの間に存在する類似点と相違点を深く探究し、同じ成分を使用してこれらを作り分けることにより、その構 造と物性特性の包括的な研究を試みた。 具体的には、ゲルとスポンジが形成する相図を作製し、両者を意図的に作り分ける高分子濃度条件を解明したことで、ゲルとスポンジを区別する条件を特定した。その結果、ゲル-スポンジ間を隔てる物性を特定し、双方の弾性率の違いや表面特性の違いが明らかとなった。さらに、開 発したゲル-スポンジの中間物質は、動物の皮下で脂肪の成長や血管構造の形成を誘導することを見出した結果、学術論文への掲載を達成した 。
また、作製したゲル・スポンジ中間物質群について、線形・非線形応答評価を行なった。回転式レオメーターを用いて線形力学応答を調査し、微小変形における弾性率と構造の相関を明らかにしつつある。非線形応答については、変形に伴う離水・体積変化に注目してデジタル画像相関法による変形速度依存性の評価をおこなった。

Strategy for Future Research Activity

ゲル-スポンジ両者を隔てる特性をさらに追求するために、形成する多孔質構造のポアサイズを意図的に制御した物質を作成し、ゲル- スポンジの弾性率、変形に伴う離水・体積変化、さらに浸透圧による熱力学的な評価を行うことで、様々な観点から統一理論の構築を目指す。 さらに、その物質での間葉系幹細胞の軟骨・脂肪・骨への分化挙動を評価することで、細胞の分化挙動に最適な多孔質構造を探索する。以上の 結果をもとに、ゲル-スポンジの構造的、および生命の物質科学的な特性の理解を追求し、双方の横断的な学理を確立する。
