Outline of Research at the Start

新規プロテインノックダウン技術であるAID2の作用原理をタンパク質構造レベルから理解し、得られた知識を反映した遺伝学的及びケミカルバイオロジー的システム改良を通して、培養細胞とマウス個体における時空間的発現制御を可能とし、さらに他の動物個体への応用可能な技術基盤を構築する。

Outline of Annual Research Achievements

昨年に引き続き、AID2による分解制御の拡張を狙い、PROTAC系のデグロンdTAGおよびBromoTagとの比較検討を行い、AID2とBromoTagを組み合わせることにより、二つのタンパク質を分解制御できることやより強力な分解誘導が可能であることを示した。実際に、AID2による分解だけでは、強い表現型変化を引き起こせない、複製因子ORC1やCDC6にダブルデグロン法を応用し、致死表現型を得ることができた。さらに、ORC1とCDC6両方をダブルデグロンで分解すると、細胞はS期を経ることなく、G2/M期に入ることがわかった。このことは、DNA複製を完全に抑制することにより、細胞周期制御からDNA複製を脱制御できることを示している。これらの技術的改良および細胞生物学的発見を、論文としてまとめ、現在リバイス論文を作成中である。AID2によるマウス作成に関しては、受精卵のROSA26座にCAG-(loxP-STOP-loxP)-OsTIR1(F74G)を挿入することを試みたが、複数回の試みにもかかわらず、ノックインマウスラインが取れなかった。そこで、ES細胞に同様のコンストラクトを導入することにした。レポーターとしてmAID-GFPをROSA26に導入したマウスは樹立できた。今後、マウスにおけるAID2システムをテストできるマウスラインの確立を目指す。脳内分解効率化を目指し、複数の新規リガンドを試したが、今の所5-Ph-IAAを超える活性を持つ化合物は取得できていない。

AID2の機能拡張は論文投稿まで進むことができた。マウスの研究に関してはmAID-GFPレポータを発現するマウスラインを樹立することができた。

Strategy for Future Research Activity

AID2の機能拡張論文の出版に力を注ぐ。すでにリバイス版作成まで来ているので、必要な実験データを追加し投稿する。マウスに関してはROSA26座にOsTIR1(F74G)を導入したマウスの樹立を進める。脳内分解用のリガンドに関しては、新規化合物のスクリーニングを継続する。
