Outline of Research at the Start

現在の創薬研究では、化合物ライブラリーのスクリーニングにより得られたリード化合物のアナログを広範に合成してそれらの活性を測定することにより構造の最適化を行う手法が主流となっている。しかし、この手法は、多大な時間と労力、費用を要することから、より効率的に医薬品候補化合物を創製する手法が望まれている。本研究では、標的タンパク質自身に阻害薬を合成させる手法である「標的誘導型合成」により、より簡便かつ迅速に、革新的医薬品候補化合物を創製する。

Outline of Final Research Achievements

In this study, we aimed to find drug candidates more easily and rapidly by "target-guided synthesis", a method that allows target proteins themselves to synthesize their inhibitors. Specifically, we performed (1) identification of mechanism-based inhibitors , (2) invitro in situ click chemistry, and (3) in cell in situ click chemistry. These works showed the usefulness of "target-guided synthesis".

Academic Significance and Societal Importance of the Research Achievements

本研究では、制限の多い銅触媒アジド-アルキン環化付加反応に拘らず、様々な反応の標的誘導型合成への適用と阻害薬が存在しない疾患関連酵素への標的誘導型合成の応用を試みた。本研究の成果は、創薬研究の加速化につながり、本手法は創薬化学の新パラダイムとなり得る。本研究により、がんや神経変性疾患のようなアンメットメディカルニーズの高い疾患に対する革新的治療薬が見い出されることも期待される。
