Outline of Research at the Start

集団で行動する動物群では、同種間で社会的優劣を決める闘いが、社会的秩序を保つ上で重要な意義を持っている。我々のこれまでの研究から、手綱核から脚間核に至る2本の神経経路が、同種間での社会的な優劣を巡る闘争における勝者と敗者の決定や、脳が向ける注意の対象を自分自身とするか外界や相手とするかという二者択一的切り替え（スイッチング）の制御に深く関わっていることが明らかになってきた。本研究では、この研究をさらに進め、手綱核・脚間核経路によるスイッチングが、脳全体の神経回路の動態にどのように影響を与え、実際の行動の違いを引き起こすのかを、関わる神経回路の全貌と作動原理を明らかにすることによって解明する。

Outline of Annual Research Achievements

昨年度の研究で、脚間核に順行的に継シナプス的に接続する神経細胞を標識できるVesicular Stomatitis Virus を注入し、脚間核と直接または間接的に接続する脳部位を同定することができた。今年度は、この実験を繰り返し、データを蓄積し、論文の執筆を開始した。さらに、脚間核の出力部位の神経細胞群を分子マーカーを使って分類するために、被蓋部分を１０X社のChromiumで１細胞mRNA発現プロファイリングを行った。
仮想空間で２匹の魚が闘争できる実験システムを確立し、正常の２匹の魚の組み合わせと、正常魚と手綱核・脚間核経路のうちで勝者の回路が不活化された魚という組み合わせえ、闘争を行い、結果を解析している。
仮想空間の中で闘争中の魚の背側被蓋部の神経活動を観察するためのグリンレンズを利用した観察システムの構築を試み、現在改良を進めている。

Reason

昨年度の研究で、脚間核に順行的に継シナプス的に接続する神経細胞を標識できるVesicular Stomatitis Virus を注入し、脚間核と直接または間接的に接続する脳部位を同定することができた。今年度は、この実験を繰り返し、データを蓄積し、論文の執筆を開始した。さらに、脚間核の出力部位の神経細胞群を分子マーカーを使って分類するために、被蓋部分を１０X社のChromiumで１細胞mRNA発現プロファイリングを行った。

Strategy for Future Research Activity

仮想空間で２匹の魚が闘争できる実験システムを使って、闘争中の魚の背側被蓋核などでの神経活動を、グリンレンズ等を使ったイマージングシステムで観察し、この部位が、内感覚受容の予測誤差に関わるかどうか等の仮説の検証を行う。
