| Outline of Research at the Start |
本研究では、ヒト胎盤幹(TS)細胞と母体細胞を共培養し、子宮内環境を高度に模倣した世界初の三次元オルガノイドモデルを作製し、母体-胎盤間のシグナル相互作用について学術的な理解を深める。また、妊娠高血圧症候群(HDP)を対象とした疾患TSオルガノイドを作製し、多階層オミックスと細胞特性の解析により、エピゲノム変異と母体とのシグナル分子機構について検討する。その際、少数細胞のエピゲノム解析を可能にする高精度、高感度の技術を応用する。さらに、多数例の蓄積したHDP疾患エピゲノムデータを、機械学習・数理モデリングに適用することで、疾患予測モデルを構築し、診断や予防に役立てる。

| Outline of Final Research Achievements |
By co-culturing of human trophoblast stem cells or blastocyst-like structures (blastoids) with endometrial cells, we created the placental/maternal organoid model that highly mimics the intrauterine environment of all the world's first. Using this model, we understood the signal transduction mechanism between mother and fetus. In addition, we created a placenta model derived from hypertension disorder pregnancy (HDP) and analyze its cell characteristics. This model and multilevel omics analysis will be applied to search for disease-causing genes.

| Academic Significance and Societal Importance of the Research Achievements |
ヒト胎盤発生過程において、母体との普遍的な相互作用を示す着床モデルを作製した。しかし、このモデルには、着床と密接に関連する免疫細胞が欠けている。そこで、私達は、前年度申請を行い、基礎的な研究を継続することを計画した。また臨床では、疾患由来TS細胞モデルを用い、周産期疾患(特にHDP)の予防や診断のガイドライン作成、さらに、医薬品産業や環境モニタリング装置の開発等に広く貢献することが十分期待できる。また、その波及効果は、発生学・幹細胞生物学などの基礎研究分野だけでなく、女性活躍社会に関わる諸制度や国や企業の少子化政策にも、重大なインパクトを及ぼす。
