Outline of Research at the Start

トカラ海峡は海洋生物の分布境界となっている。何が障害となって生物はトカラ海峡を越えられないのか? 本課題では、研究船で収集した外洋の水に環境 DNA 解析を適用し、トカラ海峡周辺域に生息する生物の正確かつ網羅的な分布マップを作成する。浮遊幼生期間の長さや産卵場の場所を含めた生物学的特徴とともに物理環境データを比較し、トカラ境界を越えるには何が必要か考察する。得られた亜熱帯域の分布マップは、漁業管理に役立つだけでなく、亜熱帯化する本州沿岸域の生態の未来を予測するのに、極めて重要な情報を提供する。

Outline of Annual Research Achievements

研究の目的:研究船で収集した外洋の水に環境 DNA 解析を適用し、トカラ海峡周辺域に生息する生物の正確かつ網羅的な分布マップを作成する。
研究実施計画:(A) 外洋での環境 DNA 採集。(B) 高精度な種判別ツールの開発。(C) 生物分布マップの作成。(D) 環境 RNA による産卵場の特定。
研究実施状況:(A) 申請者と分担者の新里、伊藤は、環境 DNA をトカラ海峡周辺域で採集している。(B) 申請者は、高精度な種判別ツール phyloSCOPE を作成し、論文として投稿した。現在、レビューワーコメントに従ってツールと論文の改訂を行っている。(B, C) 申請者は、このツールを利用して、生物分布マップの作成にも着手している。 (B, C)。サンゴを検出する環境 DNA プライマーを作成した (D) 分担者の兵藤は、環境 RNAプライマーの作成に取り組んでいる。

Reason

系統樹を推定して環境 DNA の種判定を行う phyloSCOPE (https://github.com/jun-inoue/phyloSCOPE) を論文としてまとめたため。トカラ海峡の生物境界線を検出するには、通常の環境 DNA 解析では困難である。通常の解析は配列類似性に基づいて、それぞれのサンプルに含まれる種組成推定し、トカラ海峡の両側で比較することになるが、以下が問題であった:(1) 種判定の精度が悪い。(2) 種組成の比較では検出能力が低い。 そこで本研究では、phyloSCOPEを作成した。
phyloSCOPE は上記 2 つの問題を解決するため、本研究課題に必須のウェブツールであり、他の環境 DNA 解析にも大きな貢献をすると期待できる。

Strategy for Future Research Activity

種判別ウェブツール phyloSCOPE 論文の改訂を行い、雑誌から正式に出版する。さらに phyloSCOPE を用いて、生物分布マップに必要な精度の高い種判別データを完成させる。
