Outline of Research at the Start

本研究では、光遺伝学を支えるロドプシンの作動メカニズムを、構造生物学的、分光学的、生化学的、電気生理学的手法を駆使して解明し、古くから知られているロドプシンに新しい描像を確立する。具体的に、動物ロドプシンの研究では色覚視物質の立体構造決定を試みる。微生物ロドプシンの研究では、我々が次々に発見したロドプシンの新しい機能が生まれる要因を明らかにする。ヘリオロドプシンの研究では、機能を解明するとともに、機能を生み出すメカニズムを明らかにする。

Outline of Annual Research Achievements

チャネルロドプシンをツールとして2005年に始まった光遺伝学は、脳だけでなく光による生命機能の幅広い操作を実現し、生命科学全般を革新する技術として大きな期待を集めている。ロドプシンは光遺伝学を支える標準ツールとして使われているが、私たちはこれまで、種々の新規微生物ロドプシンを発見・創成するとともに、動物ロドプシンである色覚視物質の赤外分光を用いた構造研究を世界に先駆けて行ってきた。また2018年には第三のロドプシンとも言うべきヘリオロドプシンの存在を明らかにした。
本研究では、光遺伝学を支えるロドプシンの作動メカニズムを、分光学、構造生物学、生化学・分子生物学、電気生理学を用いて明らかにする。具体的に、動物ロドプシンの研究では、色覚視物質の立体構造決定を試みる。タイプ1微生物ロドプシンの研究では、我々が次々に発見したロドプシンの新しい機能が生まれる要因を明らかにする。ヘリオロドプシンの研究では、その機能を解明するとともに、機能を生み出すメカニズムを明らかにする。本研究で対象とする様々なロドプシンは、大腸菌、酵母菌、昆虫細胞、哺乳類細胞などを用いて発現・精製する一方、必要に応じて生細胞での研究も行う。我々の学術的独自性をもたらしたのが赤外分光であり、本研究においても中心的な位置を占める。赤外分光などの分光解析に加えて、X線結晶構造解析・クライオ電顕などの構造解析、電気生理学によるイオン輸送解析などを様々なロドプシンに適用することで、作動メカニズムの解明を目指す。
以上のような3つの挑戦により、古くから知られているロドプシンに新しい描像を確立する。

Current Status of Research Progress

1: Research has progressed more than it was originally planned.

Reason

動物ロドプシン研究について、熱安定性の高いサル緑視物質変異体を対象とした構造解析を進めた結果、クライオ電顕による構造決定の可能性を持ったコンストラクトを得ることができた。一方、低温分光解析により、高度に発達した目をもつハコクラゲのロドプシンが、独自の進化により特異な静電相互作用を行うメカニズムを明らかにした。
タイプ1微生物ロドプシン研究について、これまで同様、着実に研究成果を発信することができた。具体的に、レチナールだけでなくカロテノイドを結合するプロトンポンプロドプシンやカリウムイオンを選択的に輸送するチャネルロドプシンに対して、精緻な赤外分光を用いた振動解析によりメカニズムの解明に貢献した。これらの研究成果は、それぞれ国際共同研究論文としてNatureおよびCellというトップジャーナルに掲載されている。さらに光感度の高いチャネルロドプシンGtCCR4の電気生理学実験の結果を論文発表するとともに、神取研がオリジナリティを有するロドプシン(GtCCR4, KnChR,キメラロドプシンなど)をゼブラフィッシュやマウスに適用した光遺伝学実験の成果を論文発表した。これらのロドプシンは光遺伝学ツールや視覚再生ツールとして期待が高い。実際に波及効果として特筆される光遺伝学的視覚再生に関わる2つのベンチャー企業は順調に開発を進め、その1つは大手製薬会社である第一三共に買い取られることになった。
ヘリオロドプシン研究について、イオン輸送性を持たないヘリオロドプシンが亜鉛を特異的に結合する部位を実験的に同定することに成功し、その構造様式を明らかにした。さらにこれまで未知であったシッフ塩基からのプロトン受容体が内部に結合した水分子クラスターであることを見出すことができた。

Strategy for Future Research Activity

進捗状況に記載の通り、動物ロドプシン、タイプ1ロドプシン、ヘリオロドプシンのそれぞれにおいて、順調に研究が進展している。今後、全く予見していなかった新たな展開にも対応して研究を進めたい。
動物ロドプシン研究においては、クライオ電顕による構造決定の可能性を持ったサル緑視物質変異体のコンストラクトを得ることができたため、特別推進研究の期間内に、色覚視物質の立体構造を決定できるようベストを尽くしたい。一方、クラゲやサンゴのロドプシンなど、動物ロドプシンにおいても新たな発見が相次いでいる。これら動物由来のロドプシンにおいても、大阪公大との共同研究を軸として構造ダイナミクスの解析を進める。さらにGタンパク質共役型受容体(GPCR)については関西医大との共同研究を軸として進める。
タイプ1微生物ロドプシン研究においては、引き続き様々な新規ロドプシンを研究対象として進める。特に、近赤外領域に吸収を持つベストロドプシンや酵素ロドプシンの光異性化特性の解明は、中間評価においても指摘された最重要課題の1つである。変異体やレチナールアナログなどを駆使して解明を目指す。またカルシウムイオンの結合などユニークな性質を持ったTATロドプシンのメカニズム解明も目指す。さらに応用研究である光遺伝学的視覚再生の鍵として注目される光感度を決定する要因を、GtCCR4やKnChRなど新規チャネルロドプシンの研究により明らかにしたい。
ヘリオロドプシンにおいては、イオン輸送機能を見出したV2HeR3のX線結晶構造解析を予定している。動物ロドプシンである色覚視物質の立体構造解析が、X線結晶構造解析からクライオ電顕にシフトした一方、本研究により新たに導入した結晶化用の備品を有効に活用することができる。構造を基盤としてメカニズムを詳細に解析するとともに、特徴的な分子特性をさらに深めて研究する。
