Outline of Research at the Start

現在,子どもの発達・子育てにまつわる問題が深刻化している.その一因として,親子の心身状態のシステムが不均衡であることによる相互作用の困難さがある.本研究は,養育者が育児ストレスを顕在化させやすい生後半年~1年半の乳児とその養育者を対象に,相互作用の多様性が創発する機構・機序,生物学的基盤を解明する.個体内・間で生じる身体生理の動態(リズム生成と同期)を可視化し,情報の受容―生成―伝達連鎖の因果性や,各個体の形質(身体内部状態・心的機能)との関連を明らかにする.最終的に,個々の相互作用を予測するモデルを構築し,両者の動態を安定した状態へ導き,育児効力感を高める個別型の育児支援指針を創出する.

Outline of Annual Research Achievements

新型コロナウイルス感染症の世界的拡大を受け、対面での調査や国内外の共同研究拠点との直接交流が不可能となり、研究計画の遂行は遅延せざるを得なくなった。しかし、代替となる非接触調査を検討、計画、実施したところ、想定をはるかに超える成果を得ることができた。現時点で、当初計画にある対面調査も再開できており、最終的な目標達成への影響はないと見込める。
以下、現時点で得た特筆すべき成果を3点挙げる。
(1)生後早期のヒトの迷走神経活動は、腸内細菌叢と精神・行動発達と関連することを初めて実証した。迷走神経活動は、菌叢のα多様性と正の相関を、不快な情動表出とは負の相関を示した。また、子の自律神経系・認知発達のリスクに、母親の自律神経系や精神機能が密に関連することを実証した。「腸内細菌叢-自律神経系-精神・行動」の個体「内・間」の発達的関連を示した研究は世界初である。
(2)マウスの妊娠末期~出産後の全期間の自律神経系の時系列データを初めて取得した。また、マウスの腸内細菌叢を操作すると母性行動が変化する知見も初めて得た。母マウスが仔マウスと再会すると涙液量を増やすことを見出し、その機序解明にも成功した。
(3)日常生活場面を重視した基礎研究を推進するため、3日間連続でヒト母子の自律神経系の時系列データを収集した。(クロス)リカレンスプロット解析を行ったところ、自律神経系の非線形性を失うことなく、個体内の概日リズムや個体間の相関、さらには母親の主観的ストレスを予測できた。ヒトの精神機能を予測するモデルの構築は世界的に目覚ましい勢いで進んでいるが、この成果はその最先端に位置づくにとどまらず、ヒトの育ちを支援するイノベーションへと活かされる画期的な成果といえる。

2: Research has progressed on the whole more than it was originally planned.

Reason

新型コロナウイルス感染症の世界的流行と拡大により、とくにヒトを対象とした対面での調査が長期間不可能となった。また、国内外のネットワーク拠点との直接交流を実施することも困難となったり、当初計画の見直しを迫られることとなった。また、ヒトを対象とした調査実施に不可欠となる実験装置(海外企業からの輸入品)や解析用PC・ソフトウエアが、供給の減少により納品が遅れたことも、計画の見直しに大きく影響した。

他方、本研究の実施期間を無駄に費やさないために、当初計画に代替する非接触調査を実施することとした。その成果は想定をはるかに超える、当該分野の最先端の知見を得ることができている。当初予定していたヒトを対象とした対面調査についても、現時点で再開することができており、加速的に研究を推進しておる。最終的な目標達成への影響はないことが見込める。

Strategy for Future Research Activity

【課題①】ヒト研究では、対面調査の推進に注力する。縦断的にデータを収集し、時系列解析手法を用いて、母子の自律神経系の同期パターンや精神機能・認知発達との関連について検証を進める。「腸内細菌叢-自律神経系-行動・精神」の関連が個体「内・間」でどのように推移していくかについても明らかにする。マウス研究では、妊娠期間から授乳期間における母仔の相互作用時の行動および自律神経系の時系列データをすべて取得し、両者の関係性の多様性創発の機序およびその関連要因を明らかにする。育児を開始した母マウスにストレスを与え、自律神経系、母性行動、母子間の関係性にどのような変容をもたらすかを検証する。

【課題②】ヒト研究では、自律神経系データの相互作用解析を精緻化し、その成果を学術論文にまとめる。深層ニューラルネットワークを用いて、(クロス)リカレンスプロットの画像から母子の精神機能や行動特性を直接、推定できるかを検証する。自己組織化マップを用いた推定から、深層ニューラルネットワークを用いた推定に拡張し、相互作用の動態のさらに精緻な理解を目指す。マウスで得られたデータについては、ヒトの母性行動、出産後うつ症状などと関連があった菌叢との比較検討を実施し、母性行動にかかわる細菌叢の同定を進める。また、オキシトシンを人為的に操作したマウスを用いて、母性行動の時系列変化との関連を実証する。

【課題③】ヒトとマウスのデータをもとに、相互作用時の母子(仔)の個体「内・間」の感覚運動信号や生理信号を再現・予測することのできる神経回路モデルの開発に着手する。神経回路モデルを用いた構成的アプローチと、生体データに基づく解析的アプローチの成果を統合し、養育個体-乳幼児の相互作用についてシステム的に理解する。最終的には、得られた科学的エビデンスに依拠した多様な当事者に対応しうる「個別型」育児支援の指針提案までを目指す。
