Outline of Research at the Start

分子・細胞スケールの生物学が爆発的に発展しているが、工学でしばしば重要となるマクロな連続体スケールには連結されていない。本研究ではこのボトルネックを破壊し、「生物系アクティブマターの振る舞いを予測し制御するための移動現象論」を構築する。対象とする生物はバクテリアや微細藻類などの微生物であり、腸内フローラや赤潮など、工学や医学、生物学の重要課題に実験と数理を融合して対峙する。

Outline of Annual Research Achievements

本年度の主要な研究実績は以下の通りである。 1.ヒトやマウスの内臓は、心臓が体の左側にあるなど非対称に配置されている。この左右非対称性は、胚の段階でノード繊毛が協調して流れを作り出すことによって決定されるが、そのメカニズムは長らく未解明であった。我々はマウス胚において、光ピンセットや超解像顕微鏡など独自の先進的な光学顕微鏡を用い、繊毛運動と繊毛流れの数値シミュレーションを行うことで、この問題の解明に挑んだ。その結果、繊毛カーペットが駆動する左向きの体液の流れにより、ノードの左側の不動繊毛は腹側に曲げられ、右側の不動繊毛は背側に曲げられることが明らかとなった。さらに、不動繊毛は腹側への曲げのみに反応する「曲げられる向きを感知できるアンテナ」であり、ノードの左側のみで左側を決めるシグナルを活性化できることを明らかにした。つまり、体の左右非対称性は、ベシクルなどの物質輸送ではなく、力という運動量輸送で決まることを明らかにした。この研究成果は、総合誌で世界最高峰のScience誌に掲載され、プレスリリースされた(Katoh et al., Science, 2023)。 2.流体のジェット流が液滴に分裂する現象は、Rayleigh-Plateau不安定性として知られている。本研究では、流体を「アクティブ流体」に置き換え、この古典的な現象を調べた。その結果、アクティブ流体のジェットは不安定であり、PullerとPusherの遊泳体の間で異なる不安定モードがあることが明らかになった。これらの知見はアクティブマターの特性を理解する上で重要であり、その成果は Ishikawa et al., Phys. Rev. Fluids (2022) に掲載された。 これらの業績を含め、本年度は7編の査読付き雑誌論文と22件の国際会議論文を発表し、4件の招待講演を行った。

1: Research has progressed more than it was originally planned.

Reason

これまでに、以下の特筆すべき研究業績が上がっている。1.体の左右非対称性が繊毛カーペットの運動量輸送で決まることを解明した研究成果が、総合誌で世界最高峰のScience誌に掲載され(Katoh et al., Science, 2023)、NHK NEWS WEBやYahoo Japanニュース、Newsweek日本版、朝日新聞デジタルなどに掲載されて大きな反響を呼んだ。この論文のAltmetricsは200と高く、Top 1%論文にカウントされている(Scopus調べ)。 2.環境流れに逆らう微生物の遊泳を発見した成果はScience Advances誌に掲載され(Ohmura et al., Sci. Adv., 2021)、読売新聞などに掲載されて大きな反響を呼んだ。 当初予想していなかった程のハイインパクトな成果が次々と出ているため、「当初の計画以上に進展している」と判断した。

Strategy for Future Research Activity

次年度は、主に以下の研究課題に取り組む。 1.流れ中の腸内細菌の挙動を明らかにし、挙動を表現できる数理モデルを構築する。ゼブラフィッシュ腸内の流動や細菌挙動を可視化計測できる実験系を構築し、腸内の輸送現象を実験的に計測する。 2.多細胞動物の祖先といわれる襟べん毛虫の群体に注目し、群体としての生物学的機能と進化の利点を力学的な観点から調べる。カイメン襟細胞室のポンプ機能も調べ、襟べん毛虫の群体の機能と比較検討する。 3.バイオフィルムのような微生物の集積構造を想定し、構造内の輸送現象の数値シミュレーションを行う。 4.赤潮藻の培養を行い、日周鉛直運動を再現できる実験系を構築する。背景流れによる赤潮藻の挙動の変化を計測する。 5.精子が集団遊泳するメカニズムを解明し、集団遊泳が受精機能に及ぼす影響を検討する。また、精子の走化性の数理モデル化に取り組む。 6.微生物の遊泳を工学的に応用すべく、マイクロロボットの推進原理を開発する。
