Outline of Research at the Start

地球規模の気候変動により、土壌の水分状態が大きく変動するため、作物の生育に欠かせない土壌中の養分濃度や化学形態が激しく変化している。また過度の施肥により、土壌の酸性化が進み、作物の生育を阻害する有害金属が溶出してしまい、作物の生産性や安全性において大きな問題となっている。本研究は主に”水陸両用”のイネを用いて、土壌ミネラル環境変動に可塑的に対応する植物の輸送システムを分子レベルで解明し、将来環境変動に適応できる環境応答型作物の作出に寄与する。

Outline of Annual Research Achievements

SIET4は葉の表面やケイ化機動細胞へとケイ素を排出する役割を持つことを解明した。SIET4はケイ素が蓄積する葉の表皮細胞とケイ化機動細胞の隣の細胞に局在する。SIET4 遺伝子を破壊すると、ケイ素ある条件下で栽培した場合は、生育が著しく抑制され、最後は死に至った。またOsOPT7が、OsOPT7は節や葉鞘において導管から二価鉄イオンをアンローディングする役割を明らかにした。プロテオリポソームを用いた解析により、OsOPT7は二価鉄イオン(Fe2+)をプロトンとの共輸送によって細胞内に取り込む輸送活性がみられた。OsOPT7は節の肥大維管束木部柔細胞や葉鞘の維管束柔細胞などに局在し、鉄欠乏条件下で発現が増加した。遺伝子発現抑制株では、新葉や穂への鉄分配が減少し、また葉鞘組織への鉄の分配も減少した。そのほか、イネ葉鞘と葉身間のマンガン分配に必要な輸送体OsNramp5、種子へのカルシム集積に関与する輸送体K5.2等の機能を解明した。
ミネラル環境応答シグナル因子を同定しるために、レーザー彫刻機とデジタルマイクロスコープを用いてDIYインセクトレーザー装置を開発し、イネの篩管液の採取が可能になった。ウエスタン解析やプロテオーム解析によって、ケイ素欠乏条件のイネ篩管液から、ケイ素吸収を制御する長距離シグナルタンパク質であるSSSを検出した。またCo-IPの手法を用いてこのタンパク質と複合体を形成するパートナーの探索を試みた。その結果、転写活性があり、一過性発現の実験系においてはケイ酸輸送体Lsi1の転写を負に制御する因子を単離した。
クライオ電子顕微鏡を用いてマンガン輸送体の立体構造を高分解能で解析した。マンガン輸送体について、基質の結合していない状態、マンガン基質が結合した状態、カドミウム基質が結合した状態の3つの立体構造を比較して、基質の選択に重要なアミノ酸残基の候補を明らかにした。
