Outline of Research at the Start

本研究の目的は免疫学の立場から、獲得免疫系による中枢神経機能の修復機構の解明をめざすことである。これまで脳内ではミクログリアが免疫細胞として解析されてきたものの、獲得免疫を担うT細胞、B細胞などのリンパ球はようやくその重要性が認識されつつある。しかし、その分子細胞レベルでの意義の解明は極めて遅れている。本研究では特に脳梗塞のような脳損傷やアルツハイマー病などの神経変性疾患のような脳内炎症が関与する神経傷害とその修復過程での獲得免疫系の意義を明らかにする。多彩な免疫細胞―神経細胞の相互作用を解析することで中枢神経組織の組織修復機構の解明およびその異常から生じる病態発生の理解をめざす。

Outline of Annual Research Achievements

免疫系細胞が神経系細胞を巻き込んで引き起こす「脳内炎症反応」は短期的には神経細胞にとって悪影響があるものの、炎症収束後の脳機能修復とその維持に不可欠と考えられる。本研究ではマウスモデルを中心に脳傷害時のin vivoモデルおよび一細胞RNAシークエンス(scRNAseq)解析を組み合わせることで、多様な免疫神経細胞相互作用からなる中枢神経組織の組織修復機構の解明およびその制御不全から生じる病態発生の理解をめざす。本年度はアミロイドβが蓄積しやすいAPPノックイン(APP-KI)マウスを用い、Aβ蓄積におけるT細胞の役割を検討した。T細胞を特異的に欠くAPP/CD3eKOマウスでは脳内Aβ凝集が変化し、特に早期病態においてはAβ凝集が減少することを確認した。この現象にCD8陽性T細胞が関与することを明らかにした。脳内免疫細胞のscRNA-seqでは、CD8陽性T細胞は主にケモカイン経路を介して疾患特異的ミクログリアとの細胞間相互作用を発揮し、T細胞欠損により疾患特異的ミクログリアに特徴的な遺伝子発現が亢進することから、CD8陽性T細胞は疾患特異的ミクログリアの活性化を修飾し、脳内炎症やAβ凝集に関与していると考えられた。さらにT細胞受容体レパトワ解析では、APP-KIマウス脳において抗原特異的CD8陽性T細胞のオリゴクローナルな増殖を認め、ウイルスベクターを用いてAPP-KIマウス脳に特徴的なT細胞受容体を導入したCD8陽性T細胞を移入すると脳内Aβ凝集が増加した。脳内CD8陽性T細胞は抗原特異的な活性化により、疾患特異的ミクログリアとの相互連関を介して脳内Aβ病理に関与することが示唆された。また脳内浸潤メモリーT細胞にNR4aの発現が高いことを見出した。今後NR4a欠損T細胞を用いてNR4aのAβ凝集における意義を解明する。

2: Research has progressed on the whole more than it was originally planned.

Reason

この3年間の研究ではscRNAseq解析技術を駆使して脳損傷やアルツハイマー(AD)モデルにおける脳内で各T細胞サブセットでT細胞のオリゴクローナルな増幅が認められることを明らかにし、脳抗原特異的なTCRを単離しTCR導入T細胞の移入実験をおこなっている。さらに脳梗塞慢性期に脳内で増加する自己抗体を単離しており機能面からの獲得免疫系の意義を明らかにしようとしている。このような動きは世界的にも未だほとんど報告されておらず、我々の研究が一歩リードしており、先んずることができれば学術的にも、治療診断面でも意義が大きい。ADモデルの増悪化にCD8+T細胞が関与することや、脳-腸連関による好中球の寄与を明らかにすることができた。今後その分子機構を解明するとともに、免疫細胞を標的としたアルツハイマー病の治療の可能性が示された点は画期的である。さらに本研究の過程で脳内におけるTregやメモリーT細胞にPD-1やNR4aが関与することを見出した。脳内T細胞の機能や維持機構に関する検証が進めば、NR4aが脳内T細胞を規定する新たな転写因子として注目される可能性がある。さらに認知症をはじめとする老化関連疾患とメモリーCD8+T細胞の関係の解明が進み、これらの治療にNR4aが重要な標的となる可能性がある。
このように今後進展が期待できるいくつかの重要な発見があり、本研究は順調に研究が進展していると判断している。

Strategy for Future Research Activity

1)アルツハイマー(AD)モデルで増幅されていたTCR導入CD8+T細胞のAβ蓄積に及ぼす影響を解明する。特にNR4aに焦点を絞ってAPPマウスとNR4a欠損マウスの交配を進める。
2)scRNAseqの解析からCD8+T細胞はミクログリアの分化を恒常性から病原性に転換する可能性が示された。cRNA解析を用いてCD8+T細胞がミクログリアの性質を転換させるメカニズムを明らかにする。
3)昨年度までに解析を行なった脳梗塞再発モデルにおけるTregの維持機構を明らかにする。
4)老齢脳梗塞マウスで高率で増幅されたBCRより抗体を作成し、脳梗塞マウスに投与してその意義を明らかにする。また抗原を見出す。
