Outline of Research at the Start

プログラム細胞死の分子機構を解明することは、がんの生存、増殖を理解する上で重要な研究課題である。申請者らはイリジウム錯体を基盤とするがん細胞選択的な抗がん剤の開発を通じて、カチオン性ペプチド鎖を有する錯体がプログラム細胞死の一つであるパラトーシスを誘導することを明らかにした。本研究では、光親和性標識法を用いたパラトーシス誘導金属錯体の標的分子の同定とパラトーシス誘導機構の解明を目的とする。光反応性基であるジアジリン基を導入した錯体の合成、光照射による標的分子との共有結合形成、タンパク質の分離・精製、プロテーオム解析による標的分子の同定によりパラトーシスが誘導される際の分子機構を明らかにする。

Outline of Annual Research Achievements

プログラム細胞死は細胞の生存、組織や器官形成に重要な役割を果たしており、その分子機構を解明することは新規抗がん剤の開発につながる。近年、新たなプログラム細胞死として、パラトーシス(Paraptosis)が注目されている。パラトーシスはミトコンドリア内のカルシウムイオン濃度上昇に由来する細胞質または細胞内小器官の液胞化が特徴として報告されているが、その分子機構は複雑で解明は十分に進んでいない。
申請者らは、高い発光効率を有するシクロメタレート型イリジウム(Ir)錯体にカチオン性ペプチド鎖を導入したハイブリッド錯体(IPHs)を設計、合成し、IPHsがJurkat細胞(ヒト白血病性がん細胞)の細胞死を誘導すること、死細胞中で発光するプローブとして機能することを報告した。その細胞死はパラトーシスに分類され、小胞体とミトコンドリアの膜融合に起因する直接的なカルシウムイオン輸送、濃度上昇を誘導することを見出し、従来報告されていたパラトーシスをParaptosis I、IPHsによるパラトーシスをParaptosis IIとして報告した。本年度は、光親和性標識法によるIPHsの標的分子同定、より詳細なパラトーシス誘導経路の解明を目的に研究を行った。
光照射により標的分子と共有結合を形成するジアジリン基を導入したIPHsを設計、合成、Jurkat細胞に対して光照射を行い、抽出したタンパク質をSDS-PAGEにより分析したところ、15-20 kDaにIr錯体由来の発光を有するタンパク質が検出された。次に、本タンパク質を詳細に分析するために、2次元電気泳動を行ったが、Ir錯体由来の発光は検出されなかった。この理由として、タンパク質の総量が少なかったこと、泳動ゲルの固定化に使用した酸によってIr錯体が分解したことが考えられる。今後は、上記の問題解決に努め、標的タンパク質を同定する予定である。
