Outline of Research at the Start |
The goal of this project is to develop a combined Raman & fluorescence imaging flow cytometer with cell sorting capability for the analysis and selection of microalgae that hold high potential as producers of medical bioproducts, bioplastics, and biofuel.
|
Outline of Annual Research Achievements |
The goal of this project was to establish simultaneous Raman & fluorescence cell sorting based & imaging flow cytometry. This required combined development in three fields: nonlinear spectroscopy, microfluidics, and real-time data processing and decision making. At the time of this report, I have achieved cell sorting based on analysis of broadband, fingerprint region Raman spectra at throughputs of up to ~50 cells/s and purities up to 93%. This work required constructing the cell sorter, a proof of concept sorting using the Raman signal from polymer microbeads, and sorting cells based on the Raman signal from their polysaccharide and carotenoid content. A manuscript based on the results is currently under review.
