Outline of Research at the Start

肝蛭症は人獣共通感染性の吸虫症である。アジアには単為生殖型肝蛭が分布する。単為生殖型肝蛭には祖先種と同じ2倍体に加えて、3倍体がある。3倍体は病害が強いため、被害状況の把握が強く求められているが、アジアでは人体症例の原因種すら正確に診断されていない。また、分類学的な種が未確定のため、医療・獣医療従事者に認識されていない。本研究では、定量的PCR法を開発し、単為生殖型3倍体の遺伝子型を解析する。これにより、3倍体の ① 分子学的鑑別法が世界で初めて確立され、②大規模疫学調査が実現する。さらに、遺伝子型の解析により3倍体の出現様式を解明できるため、③ 単為生殖型肝蛭の分類学的位置を決定できる。

Outline of Annual Research Achievements

リアルタイムPCR装置の変更に伴い、定量的PCRの条件検討に時間を要したが、今年度までにその問題を解決することに成功した。
具体的には、はじめに、より感度が高い定量的PCRキットに変更した。次に、抽出後のゲノムDNAを2-3倍階段希釈してから定量的PCRを実施し、増幅効率を算出し、適当な増幅効率を満たした結果を採用した。単為生殖型肝蛭はF. hepaticaとF. giganticaの交雑子孫であるが、2倍体の場合、F. hepaticaとF. giganticaのゲノムを等量ずつ有するので、両種に特異的なプライマーセットを用いると、定量的PCRのCt値の差が0になる。一方、3倍体の場合は遺伝子型に応じて、F. hepaticaもしくはF. giganticaのゲノムが1コピーずつ多くなるため、Ct値の差が1になる。適切な増幅効率を満たした反応条件で得られたCt値について、3点もしくは4点の階段希釈倍率で得られた結果を平均することで、遺伝子型の判定を最適化することができた。
この方法を用いれば、死後時間を経過してから固定されたサンプルから抽出したDNAサンプルでも判定に用いることができると考えられる。
最終年度にはこの方法をpepckとpoldを対象にしてフィールド株に適用したい。また、昨年度確立した新たな核DNAマーカーFABP typeIについても検討を進め、単為生殖型2倍体と3倍体の分布状況と遺伝子型を明らかにし、3倍体の出現様式の結論を出したい。

これまでに改善したプロトコルを用いて、遺伝子型が既知の実験室株と未知のフィールド株の両方に定量的PCRを適用し、単為生殖型肝蛭2倍体と3倍体の識別を可能にすると同時に、3倍体の出現様式を明らかにしたい。
