Outline of Research at the Start

Outline of Final Research Achievements
This study evaluated the immunogenicity of a ΔZmp1-BCG strain, in which the Mycobacterium tuberculosis virulence factor Zmp1, which isknown to suppress IL-1β production, was deleted. Mice were immunized with either ΔZmp1-BCG or wild-type BCG and subsequently challenged with M. tuberculosis via the airway. Both vaccinated groups showed reduced infiltration of inflammatory cells in the lungs compared to the unvaccinated control group. The ΔZmp1-BCG group exhibited higher expression levels of IL-6, TNF-α, and IL-17A, with a significant increase in IL-17A-producing γδT cells (γδT17). Although pulmonary bacterial loads were only moderately reduced, a significant decrease in bacterial counts was observed in the spleen. These findings suggest that ΔZmp1-BCG may suppress extrapulmonary dissemination of M. tuberculosis via γδT17-mediated immune responses and holds promise as a more effective tuberculosis vaccine candidate.

Academic Significance and Societal Importance of the Research Achievements
