Outline of Research at the Start

自閉スペクトラム症(ASD)患者は聴覚過敏を示す場合が多いが、その病態は不明である。本研究では、有力なASDリスク遺伝子の中で聴覚神経系および機能との関連が最も示唆されている分子Contactin5(CNTN5)に着目する。Cntn5全欠損およびexon4(ヒトでこのexonおよび近傍配列に異常が複数認められている)欠損マウスを用いてCntn5の機能解析をすることにより聴覚過敏の発症メカニズムを解明および治療法の開発をすることを目的とする。

Outline of Annual Research Achievements

Contactin5 (CNTN5)は、聴覚系において脳幹のグルタミン酸作動性ニューロンの発達に関与し、CNTN5 KOマウスで聴覚脳幹反応(ABR)の脳波潜時が延長していた(Toyoshima et al., Dev Biol, 2009) 。また、CNTN5および6の遺伝子変異を持つ自閉スペクトラム症 (Autism Spectrum Disorder; ASD)患者は聴覚過敏を持ち、ABRの脳波潜時の延長を認めた(Mercati et al., Mol Psychiatry, 2016)。CNTN5の全エクソンをカバーするように設計した22個のプライマーを用いて、ASD患者のDNAサンプルのexon sequenceをABI PRISM 3130 x l Genetic Analyzerで行う。ASD群で検出された変異についてTaqman法で健常コントロール群に存在するかを確認して、症例対照で頻度を比較する。それらの変異についてin silico解析でその変異の有害度を推測した。これまでに8個のミスセンス変異が同定された
が、そのうち4つ(rs778655565, rs186615197, rs771277271,rs757538847)がin silico解析(PolyPhen-2)によりDamagingと判定された。この4種の変異Cntn5-Fc融合タンパク溶液を作成し、CNTN5-FcとPTPRG(CNTN5と結合することにより神経突起伸長を促進)のcell surface binding assayを実施し、いずれの変異型CNTN5-Fcも野生型と比べてPTPRGとの結合が弱いことを示した。
またASDへの関連が示唆されているMAP4Kの阻害が神経発達に負の影響(神経突起長の減少)を与えるという研究結果をハイコンテントスクリーニングを用いて示し、Mol brain誌に採択された (Lasham et al., 2023)。

Strategy for Future Research Activity

CNTN5のエクソンシークエンスにより同定されたミスセンス変異のうち、in silico解析により有害と判定された変異をプラスミドベクターを用いて培養神経細胞に導入して、神経突起長解析では3日間、シナプス解析では2週間、培養したのちに固定して、染色(神経突起マーカー:MAP2、前シナプスマーカー:Synaprophysin1、後シナプスマーカー:SHANK2)、細胞イメージアナライザー(CellInsight CX5)で神経突起長およびシナプス数を自動計測し、コントロールとそれぞれの濃度の平均値をt検定で比較。多重比較の補正(Bonferroni法)を行う。
