Outline of Research at the Start

本研究では,パターン認識受容体の一種であるcGAS (cyclic GMP-AMP synthase)が介在する放射線応答制御機構を明らかにする。加えて,癌治療において悪影響となる細胞老化随伴分泌現象(炎症因子等の分泌現象)とcGASとの関わりに着目し,cGAS刺激因子と放射線の併用時における細胞老化随伴分泌現象ならびに分泌因子による癌病態及び正常細胞への影響を明らかにし,効果的な癌放射線治療戦略の構築に役立てる。

Outline of Annual Research Achievements

cGAS (cyclic GMP-AMP synthase) は病原体関連分子を認識するパターン認識受容体の一種で,加えて近年では,免疫細胞内のcGASの活性化が抗腫瘍免疫誘導に重要であることが示されている。一方,癌細胞内でのcGASの活性化が癌治療効果に及ぼす影響に関する報告は限られている。研究代表者は公共データベースを用いた解析によりcGASの発現と頭頸部扁平上皮癌患者の予後不良等との関連性を見出すとともに,in vitroにおいてcGASが頭頸部扁平上皮癌細胞の放射線抵抗性に関与することを見出したが,そのメカニズムは未解明である。本研究では,cGAS刺激因子と放射線の併用による抗腫瘍免疫活性を兼ね備えた効果的な癌放射線治療戦略に繋げるための基盤研究として,放射線の抗癌効果の妨げとなるcGASが介在する癌細胞の放射線抵抗性機構を解明する。
1,2年度では,癌細胞の放射線応答制御におけるcGASの役割とその機構解明に取り組み,以下のことを明らかにした。①cGASが頭頸部扁平上皮癌細胞の放射線抵抗性に寄与する。②cGASは癌細胞株の種類に応じて放射線誘発分裂期崩壊または放射線誘発アポトーシス・細胞老化を制御し,放射線抵抗性を制御する。また,この細胞株依存的な放射線応答性の違いは照射後のp21の応答性の違いに起因する。
本年度はcGASが関わる細胞老化と細胞老化随伴分泌現象に着目し,以下のことを明らかにした。ヒト血管内皮細胞における放射線誘発細胞老化にcGASとは別のパターン認識受容体であるRIG-I様受容体が関わっていることを明らかにした。また,分泌現象による癌細胞への影響を共培養実験で検証したところ,老化した正常細胞は非照射及び照射癌細胞のクローン形成能を促進させることが明らかとなった。

細胞老化随伴分泌現象による癌細胞への影響解明に取り組み,重要な知見を得たため。また,ヒト血管内皮細胞における放射線誘発細胞老化にcGASとは別のパターン認識受容体であるRIG-I様受容体が関わっていることを明らかにし,その成果を国際学術誌『Biomedical Reports』で発表したため。

Strategy for Future Research Activity

次年度は,cGAS及び細胞老化が関わる放射線応答制御機構をより詳細に解析し,癌細胞に対する放射線の致死効果増強及び正常組織の障害改善に役立て,放射線療法の効果向上に繋げる。
