Outline of Research at the Start

成長障害の大半はいまだに原因が不明である。成長は、成長軟骨帯の伸長によって起こり、成長促進因子としてインスリン様成長因子-1(IGF-1)が最も有名である。ところが、IGF-1が成長軟骨で、どのような分子基盤を介して伸長作用を発揮するのか、いまだに不明である。そこで、IGF-1受容体(IGF1R)異常症患者由来のiPS細胞より誘導した軟骨細胞を用いて、軟骨におけるIGF-1の作用機序および成長障害の分子基盤を明らかにすることで、新規治療の開発に結び付けたい。

Outline of Annual Research Achievements

小児内分泌診療において最も重要なテーマの一つは成長障害である。成長は成長軟骨帯の伸長によって起こり、成長促進因子として最も古くから研究され、臨床応用もされているのは、成長ホルモン(GH)/インスリン様成長因子-1(IGF-1)である。ところが、GH/IGF-1がどのような分子基盤を介して成長軟骨帯の伸長を促進するのか未だに不明である。本研究では、ヒトiPS細胞を軟骨に分化誘導し、IGF-1受容体の下流の分子を網羅的に解析することで、IGF-1によるヒト成長軟骨伸長作用の分子基盤の解明を目指す。
令和3年度からヒトiPS細胞からヒト軟骨細胞を分化誘導する培養系の最適化を目指して試行錯誤を繰り返した結果、当研究室において最も安定してiPS細胞から軟骨細胞に分化誘導するには、Pretemerら(Stem Cell Rep 2021)らの方法が最適であると判断した。軟骨細胞は分化誘導後も静止軟骨細胞、増殖軟骨細胞、肥大軟骨細胞と分化し続けるため、現在はそれぞれの分化段階におけるIGF-1シグナルを解析している。

3: Progress in research has been slightly delayed.

Reason

コロナ禍のため臨床業務が増加し、令和3年度、4年度は実験・研究時間の確保が困難であったが、令和5年度になり、新型コロナウイルス感染症が5類感染症に移行したため、コロナ対応に要するエフォートは減少し、臨床業務はコロナ以前に近い状況にもどった。実験・研究にまとまった時間を割くことができるようになり、令和3年度、4年度と比較して比較的スムーズに実験を重ねることができたが、まだ2年分の遅れは取りもどせてはいない。

Strategy for Future Research Activity

軟骨細胞は分化誘導後も静止軟骨細胞、増殖軟骨細胞、肥大軟骨細胞と分化し続けるため、現在は培養期間、細胞形態、軟骨分化マーカーの発現からそれぞれの分化段階において細胞の分化が最も均一な条件の探索、および各々の条件におけるIGF-1シグナルを解析を行っている。これらのデータを統合して静止軟骨細胞、増殖軟骨細胞、および可能であれば肥大軟骨細胞においてRNA-Seq解析を行い、IGF-1添加の有無によるmRNAの発現変化を解析する。
