Outline of Research at the Start

腸管生物時計は栄養素の吸収機構を制御し、その破綻はグルコースを含めた栄養素の吸収障害を引き起こす。一方、妊娠母体の生物時計の破綻は子宮内発育遅延と関連することが報告されている。子宮内発育遅延児は、胎児プログラミング機構を介して成人期の耐糖能異常と関係するため、妊娠母体の生物時計の破綻は出生児の成人期の耐糖能異常の原因になりうる。しかし、その詳細な分子機構は不明である。本研究では、腸管生物時計の破綻はグルコース吸収低下により低栄養の原因となり、妊娠中には胎児低栄養を介して、出生児の成人期の耐糖能異常の原因となるという仮説の検証を行う。

Outline of Annual Research Achievements

生物時計は地球の自転に伴う環境のサイクルと体内のサイクルを同調させるシステムである。生物時計は種々の代謝恒常性維持制御に関わっているが、腸管生物
時計は栄養素の吸収機構を制御し、その破綻は栄養素の吸収障害を引き起こす。一方、妊娠母体の生物時計の破綻は子宮内発育遅延と関連することが報告されて
いる。子宮内発育遅延児は、胎児プログラミング機構を介して成人期の耐糖能異常と関係するため、妊娠母体の生物時計の破綻は出生児の成人期の耐糖能異常の
原因になりうる。しかし、その詳細な分子機構は不明である。そこで、本研究では腸管生物時計に注目し、①腸管生物時計の破綻はグルコース吸収低下を引き起
こし低栄養の原因になる、②妊娠母体の腸管生物時計の破綻は胎児低栄養を引き起こし、胎児プログラミング機構を介して、出生児の成人期における耐糖能異常
の原因になるという2つの仮説を、腸管特異的に時計遺伝子Bmal1を欠損するマウスを用いて時間栄養学の観点から検証することが本研究の目的である。初年度は、腸管特異的Bmal1欠損マウスを用いて、生物時計によるブドウ糖吸収制御機構の解析を行った。ブドウ糖吸収に関わるSGLT1にはリズム性の発現があり、BMAL1/CLOCKによる制御を受けていることを見出した。さらに、腸管特異的Bmal1欠損マウスではブドウ糖吸収が時間依存性に低下していることを見出した。現在、腸管特異的Bmal1欠損母マウス由来仔マウスとコントロール母由来仔マウスの間における糖代謝の差異を検討している。腸管特異的Bmal1欠損母マウス由来仔マウスでは、対象群と比べ体重増加に変化を認めなかったが、耐糖能が悪化していることを見出した。

Reason

腸管特異的Bmal1欠損母マウス由来仔マウスとコントロール母由来仔マウスの間における糖代謝の差異を検討している。腸管特異的Bmal1欠損母マウス由来仔マウスでは、対象群と比べ体重増加に変化を認めなかったが、耐糖能が悪化していることを見出したが、表現型が軽微であり、その確定に時間を要したため。

Strategy for Future Research Activity

腸管生物時計によるブドウ糖吸収異常が全身の糖代謝維持機構に与える影響の検討をさらに行う。具体的には、高脂肪食摂餌などの環境負荷による表現型の変化を検討する。
さらに、糖代謝に関わる組織(肝臓、骨格筋、脂肪組織ばど)における遺伝子発現の違いなどを、RNAシーケンス解析などで確認し、腸管生物時計の破綻によるブドウ糖吸収不全によるDOHaDの分子機序を解明していく。
