Outline of Research at the Start

造血不全症の代表疾患の一つである再生不良性貧血は、骨髄の造血幹細胞の減少と脂肪髄を特徴とする難治性疾患の一つである。これまでに申請者らのグループは、造血幹細胞と造血幹細胞を支持する造血微小環境を構成する間葉系幹細胞の両者における転写因子GATA-2の発現低下が、造血細胞の減少と脂肪髄という二つの異常形質と深く関わることを見出してきた。さらに間葉系幹細胞におけるGATA-2の発現低下により造血支持能が低下する点も明らかとした。しかしながら、造血幹細胞の質的異常が造血微小環境に与える影響については不明である。本研究ではこの点を明らかとすることを目的とした。

Outline of Annual Research Achievements

造血不全症の代表疾患の一つである再生不良性貧血は、骨髄の造血幹細胞の減少と脂肪髄を特徴とする難治性疾患の一つである。これまでに申請者らのグループは、造血幹細胞と造血微小環境を構成する間葉系幹細胞の両者における転写因子GATA-2の発現低下が、造血細胞の減少と脂肪髄という二つの異常形質と深く関わることを見出してきた。しかしながら、造血幹細胞の質的異常が造血微小環境に与える影響については不明である。
本研究においては、造血幹細胞特異的にGata2をノックアウトする方法として、Cre-loxPシステムを用いる。まずGATA2f/fマウスと,Rosa26領域にエストロゲンレセプターとCre融合蛋白をコードする配列が導入されたER-Creマウスを交配することにより,タモキシフェン投与にてGATA2がノックアウトされる条件付きノックアウトマウスを作製した。GATA2f/f/ER-Creマウスにタモキシフェンを腹腔内投与した後、GATA2のノックアウト効率を定性PCRによるゲノムDNAの組み換え
や定量PCRによるmRNAの発現量にて確認した。同マウスの骨髄を評価したところ、Gata2ノックアウトマウスにおいては骨髄造血細胞の著明な減少を認めた。しかし、本マウスはGata2を発現するすべての細胞においてGata2をノックアウトするため、マウス造血幹細胞において高発現していると考えられている、Fgd5遺伝子のプロモーター領域の制御下にCreを発現させるマウスを用いて、造血幹細胞特異的にGata2をノックアウトさせるマウス(Fgd5-Cre-ER;Gata2flox/floxマウス)
の確立を試みている。

3: Progress in research has been slightly delayed.

Reason

GATA2f/f/ER-Creマウスにタモキシフェンを腹腔内投与した後、同マウスの骨髄を評価したところ、Gata2ノックアウトマウスにおいては骨髄造血細胞の著明な減少を確認できたが、次のステップである、造血幹細胞特異的にGata2をノックアウトさせるマウス(Fgd5-Cre-ER; Gata2flox/floxマウス)の確立に関しては、マウスFgd5遺伝子とマウスGata2遺伝子が同一染色体上で近接しており、Fgd5-CreとGata2floxを1つの染色体座に同時に発現させる点で苦慮している。

Strategy for Future Research Activity

マウスFgd5遺伝子とマウスGata2遺伝子が同一染色体上で近接しているものの、ある確率で組み換えが起こり同一染色体上に2つの遺伝子座が存在することもありうるため、交配を継続している状況である。またマウスの系の樹立が難しい場合は、ヒト臍帯血由来造血幹細胞の研究も検討する予定である。
