Outline of Research at the Start

本研究は解離病態におけるTreg細胞の役割を明らかにし,Treg細胞の制御メカニズムをSykに着目して解明することである。近年の研究で、解離病態における組織破壊メカニズムは解明されつつある。しかし炎症応答の恒常性破綻メカニズムは未開拓の分野である。Treg細胞は、過度な炎症を抑制して生体の恒常性を保っている。本研究では、免疫制御分子SykがTreg細胞活性を維持し解離を抑制するとの独自の知見に基づき着想された。研究成果を解離病態に位置付けることで、Treg細胞を中心とした生体恒常性の破綻機構が解明される。さらにSykとTreg細胞の関連が明らかになることで生命現象に新たな視点が得られる。

Outline of Final Research Achievements

We investigated the spatial distribution of inflammatory cell subsets in human aortic dissection tissues and identified two distinct cellular clusters: one composed of B cells and helper B cell subsets, and the other of M1 and M2 macrophages as well as neutrophils. These clusters were found to be interconnected through B cells and M2 macrophages. Analysis using a murine model of aortic dissection suggested that the smooth muscle cell signaling molecule Syk regulates anti-inflammatory responses, including those mediated by Treg cells, thereby suppressing dissection. Transcriptomic dataset analysis revealed regulatory relationships among genes associated with dissection, indicating that the coordinated regulation of genes involved in cell proliferation, inflammatory responses, cell motility, morphogenesis, and sensory organ development contributes to the pathogenesis of aortic dissection.

Academic Significance and Societal Importance of the Research Achievements

大動脈解離において炎症応答が重要であることが指摘されているが、解離分子病態の中で炎症応答が占める位置や制御メカニズムは不明である。今回の研究では、大動脈解離病態における炎症細胞の相互作用、炎症シグナル分子Sykの関与、全遺伝子発現における炎症関連遺伝子群の位置付けを明らかにした。解離病態は細胞間相互作用や遺伝子間相互作用が連続的に起こるダイナミックな病態であり、その解明にはネットワークとしての挙動を解明する必要があると考えられた。この観点から診断マーカーや治療標的を同定していく必要があると思われる。
