Outline of Research at the Start

敗血症の治療では、炎症メディエーターの腫瘍壊死因子(TNF-α)の合成や誘導型一酸化窒素合成酵素(iNOS)などの発現を抑制することが重要である。我々はiNOS アンチセンス転写物(asRNA)に対して、(1)asRNAと相補的な塩基配列を持つオリゴヌクレオチド(センスオリゴ)が、肝細胞内において阻害効果を示すことや(2)肝障害敗血症モデルラットにiNOSセンスオリゴを投与すると、生存率が増加することを報告した。今回、敗血症に対する更なる効果の向上を目的として、(1)iNOSセンスオリゴの改良、(2)既存の別機序による抗炎症効果を持つ薬物との併用効果の評価を行う。

Outline of Annual Research Achievements

敗血症に対する更なる効果の向上を目的として、(1)初代培養ラット肝細胞および敗血症ラットモデルを用いたSO1の改良、(2)既存の別機序による抗炎症効果を 持つ薬物との併用効果の評価、(3) iNOSや多数のICの発現を制御する重要な遺伝子mRNAをターゲットとした新規センスオリゴを行った。
(1)昨年度行った難分解性の修飾の改良に加えて、肝臓への移行性を強化する化学修飾を行った改良型SO1を作成した。現在ラット敗血症モデルにおける効果検討を行っている。(2)ヒト敗血症領域で、汎発性血管内血液凝固症に対して使用される遺伝子組換えトロンボモジュリン製剤(rTM)とSO1の併用効果を検討し、異なる機序での効果を確認した(Richi Nakatake et al., “Combination therapy with a sense oligonucleotide to inducible nitric oxide synthase mRNA and human soluble thrombomodulin improves survival of sepsis model rats after partial hepatectomy” “Shock”, Wolters Kluwer, Vol.60, no.1 pp.84~91 (2023))。(3)新たな遺伝子(IL-1関連)を阻害するセンスオリゴを設計した。敗血症モデルラット(肝切除後敗血症モデル)にセンスオリゴ投与した効果検討を行い、有意な改善を認めた。盲腸結紮穿刺による腹膜炎敗血症モデルにおいても改善を認めている。現在ヒトへの応用を目標として、センスオリゴのヒト配列化を行っている。
