Outline of Research at the Start

神経麻痺性角膜症とは三叉神経の障害に伴って角膜上皮の創傷治癒遅延や難治性上皮欠損を発症する疾患である。
本課題では、(1) Shhとネトリン1を介した三叉神経;角膜上皮・輪部幹細胞の相互作用の分子機構を解明し、(2) Shh由来N末端ペプチド/ネトリン1由来ペプチド投与のマウス神経麻痺性角膜症に対する治療効果を明らかにする。神経麻痺性角膜症の病態解明と新規治療戦略の樹立に貢献すると期待できる。

Outline of Annual Research Achievements

角膜上皮または感覚神経特異的遺伝子改変マウスを実験に供給するために継続繁殖している。
昨年以来、感覚神経特異的Advillin-Cre/Shh floxマウスは研究遂行に十分な数が得られず、実験が遅延している。並行しているrtTA/K12-Cre/Shh floxマウスで輪部から角膜に侵入する三叉神経末梢感覚神経分布の表現型に軽度の差が見られた。しかし、コントロールマウスとrtTA/K12-Cre/Shh floxマウスでShh mRNA発現に統計学的有意差がなく、遺伝子発現が変化する状況下での研究を発案するに至った。すなわち、上皮創傷治癒の過程の両群マウスでのShh発現の変動を評価すべく、マウスを繁殖する計画とした。
完全に組織特異性を発揮するrtTA/K12-Cre/Shh floxマウスと、Advillin-Cre/Shh floxの双方を十分に確保した上で、上皮創傷治癒過程での生体反応を評価する必要があると考えるに至った。

目的の遺伝子を持ったマウス(完全に組織特異性を発揮するrtTA/K12-Cre/Shh floxマウスと、Advillin-Cre/Shh floxの双方)の十分数が得られないため。

Strategy for Future Research Activity

角膜上皮でのShh発現が完全に抑制されていないので、上皮の創傷治癒過程ので研究モデルを作成、遺伝子発現が動く場合での遺伝子改変による影響を検討しなければならないと言う認識に至った。創傷下での遺伝子発現の変化の確認がその後のペプチド治療の実験の布石となる。完全に組織特異性を発揮するrtTA/K12-Cre/Shh floxマウスと、Advillin-Cre/Shh floxの双方、が実験に使用する十分数に到達するように努める。
