Outline of Research at the Start

角膜上皮幹細胞疲弊症では幹細胞疲弊に伴う結膜および血管侵入に加えて眼表面上皮の分化異常が生じる。その結果、正常上皮が角化(皮膚化)して高度の角膜混濁をきたし、著明な視力低下を引き起こす。本研究では、角膜上皮の細胞分化を規定する転写因子ネットワークを、スーパーエンハンサーによる制御を中心に解析することで、角膜上皮細胞と表皮角化細胞(皮膚上皮細胞)の角化と非角化を制御する分子メカニズム機構を解明する。

Outline of Annual Research Achievements

以前に6つのコア転写因子(PAX6, OVOL2, KLF4, SOX9, TP63, MYC)を用いて皮膚線維芽細胞から角膜上皮細胞へのダイレクトリプログラミング(ある分化細胞から違う分化細胞の強制誘導を行う方法)に成功し、これらのコア転写因子が角膜上皮の正常分化を制御することを報告した。ドナー角膜を海外のアイバンクから研究用として入手し、ドナー角膜から角膜および結膜上皮細胞のみをディスパーゼを用いて単離し、昨年からヒト培養角膜上皮細胞およびヒト培養結膜上皮細胞の細胞老化について研究を進めている。X線照射により細胞老化を誘導した角膜上皮細胞と非老化誘導角膜上皮細胞を比較したところ、合計3295個の遺伝子の発現が異なっており、そのうち、1200個の遺伝子が上方制御され、2095個の遺伝子が下方制御されていた。一方、老化を誘導した結膜上皮細胞と非老化誘導結膜上皮細胞を比較したところ、合計2642個の遺伝子の発現が異なっており、そのうち、883個の遺伝子が上方制御され、1759個の遺伝子が下方制御されていた。さらに、老化角膜上皮細胞と老化結膜上皮細胞を比較したところ、老化結膜上皮細胞で角化関連遺伝子の発現が有意に上がっていることを見出した。さらに、スティーブンスジョンソン症候群、眼類天疱瘡といった眼表面の角化を引き起こす角膜上皮幹細胞疲弊症である、難治性眼表面疾患の眼表面組織を手術時に採取して免疫染色をおこなった。その結果、同年齢の正常結膜組織と比較して、難治性眼表面疾患の眼表面組織ではp16陽性の老化細胞が存在することがわかり、p16陽性の組織では角化関連のタンパク発現が上昇していることがわかった。このことは、老化細胞が眼表面の角化の病態に関与していることを示唆する。

Reason

これまでATAC-seqを行うことで角化の病態を解析することを試みていたが、昨年から老化細胞の検討を開始ししたところ、老化結膜上皮細胞では特に角化関連分子の発現が上昇していることがわかり、角化の病態解明に繋がる可能性が示唆されており順調に進捗している。

Strategy for Future Research Activity

X線照射だけでなく、UV照射も用いて細胞老化を誘導し、ヒト培養角膜上皮細胞およびヒト培養結膜上皮細胞の解析を行い、細胞老化と角化の観点から解析を進めることで角化と非角化を制御する分子メカニズムの解明を行う。
